5种培养基对蛋白核小球藻生长及其产物合成的影响
Effects of Five Media on the Growth and Product Synthesis of Chlorella pyrenoidosa
【摘要】 以蛋白核小球藻CP-FJ9为出发藻株,在异养培养条件下,采用SE、HA-SK、BG11、Basal和BBM 5种培养基分别对小球藻CP-FJ9进行培养.通过测定生物量、比生长速率及细胞内主要成分,比较5种培养基对小球藻CP-FJ9生长及其产物合成的影响.实验结果表明:Basal培养基更适合小球藻CP-FJ9快速培养,可获得最高生物量12.93 g·L-1和最大比生长速率0.64 d-1;而HA-SK培养基则更有利于蛋白质的合成,可获得最高蛋白质产量5.14 g·L-1.另外,在10 L发酵罐中使用HA-SK培养基异养培养蛋白核小球藻CP-FJ9,经72 h发酵后,小球藻最终生物量为20.05 g·L-1,蛋白质含量为49.06%.这表明,蛋白核小球藻CP-FJ9是一株有潜力用于开发富含蛋白质的藻种.
【Abstract】 Chlorella pyrenoidosa CP-FJ9 as the starting strain was cultured in SE, HA-SK,BG11,Basal, and BBM media respectively under heterotrophic conditions.The effects of five media on the growth and the product synthesis of C.pyrenoidosa CP-FJ9 were compared by measuring biomass, specific growth rate and main components in cells.The results showed that Basal medium was more suitable for the rapid cultivation of C.pyrenoidosa CP-FJ9,and the highest biomass(12.93 g·L-1) and maximum specific growth rate(0.64 d-1) were obtained; While HA-SK medium was more conductive to the protein synthesis of C.pyrenoidosa CP-FJ9, and the highest protein yield reached 5.14 g·L-1. In addition, this microalga heterotrophically cultivated with HA-SK medium in the 10 L fermenter attained, after 72 hours of fermentation, the final biomass of 20.05 g·L-1 with 49.06% protein content.Taken together, these results show that C.pyrenoidosa CP-FJ9 isa potential strain for the development of protein-rich algae.
