茶树TLP家族基因鉴定及其对高温干旱和炭疽菌侵染的响应

Identification of TLP gene family in Camellia sinensis and their expression pattern under high temperature, drought stress and Colletotrichum gloeosporioides infection

【摘要】 类甜蛋白(TLP)家族基因在植物抵御逆境胁迫过程中发挥着重要作用.本研究利用生物信息学方法对茶树TLP家族基因进行分析和预测,并通过qRT-PCR检测了其在炭疽菌侵染和高温干旱胁迫下的表达水平.从茶树基因组中共鉴定出31个TLP基因,命名为CsTLP1~CsTLP31.系统进化分析表明,31个TLP基因分成10个类群,其中,类群5中的成员最多,且集中在13、14号染色体上.结合基因的表达特征可知,类群5和类群6中的较多成员在逆境胁迫下呈现差异表达.qRT-PCR验证表明:在炭疽菌侵染胁迫下,CsTLP20、CsTLP21、CsTLP23、CsTLP27的表达量与对照相比显著上调;在高温干旱胁迫下,10个TLP基因的表达量与对照相比存在显著差异,其中,CsTLP20、CsTLP21、CsTLP22、CsTLP23、CsTLP28的表达量显著上调,CsTLP7、CsTLP16、CsTLP27的表达量显著下调,CsTLP15、CsTLP26的表达量在部分时间点显著上调.综合来看,CsTLP20、CsTLP21、CsTLP23的表达量在炭疽菌侵染和高温干旱胁迫下均显著上调,表明它们是TLP家族基因中参与逆境响应的重要成员.研究结果为进一步开展茶树TLP家族基因的功能验证和抗逆机理研究提供参考.

【Abstract】 Thaumatin-like protein(TLP) gene family play important roles in plant′s resistance to stresses arisen from adversity. In this study, the TLP gene family of tea plants were predicted and analyzed using bioinformatics methods, and their expression levels under Colletotrichum gloeosporioides infection, high temperature and drought stress were further examined by qRT-PCR. A total of 31 TLP genes, CsTLP1-CsTLP31, were identified from the genomic database of Tea Plant Information Archive. Phylogenetic analysis showed that the 31 TLP genes were divided into 10 groups, in which group 5 had the most members and were mainly located on chromosomes 13 and 14. According to the expression characteristics of the genes, more members of group 5 and 6 presented differential expressions under adversity. qRT-PCR verification further showed that the expressions of 4 TLP genes, including CsTLP20, CsTLP21, CsTLP23 and CsTLP27, were significantly up-regulated under C.gloeosporioides infection; under high temperature and drought condition, the expressions of 10 TLP genes were significantly different from those of the control, among which CsTLP20, CsTLP21, CsTLP22, CsTLP23 and CsTLP28 were significantly up-regulated while CsTLP7, CsTLP16 and CsTLP27 were significantly down-regulated, and CsTLP15 and CsTLP26 were significantly up-regulated at some time points. In summary, CsTLP20, CsTLP21 and CsTLP23 were significantly up-regulated under both types of stresses, indicating their key role as the TLP gene family in stress response. The study provided a reference for further functional validation of the TLP gene family of tea plants and research on the mechanism of stress resistance.
