植物炭黑(百草霜)毒理学安全评价试验
Study on toxicological safety evaluation of plant carbon(plant soot)
【摘要】 本试验旨在对植物炭黑进行毒理学安全评价。小鼠急性毒性试验(LD50测定)按植物炭黑5 000 mg/kg体重剂量经口灌胃,观察试验期内小鼠中毒症状、大体剖检及死亡情况。小鼠骨髓细胞染色体畸变试验及小鼠精子畸形试验以环磷酰胺40 mg/kg体重为阳性对照组、1%羧甲基纤维素钠为阴性对照组,植物炭黑设5 000 mg/kg体重、2 500 mg/kg体重、1 250 mg/kg体重3个剂量组,经口灌胃,观察试验小鼠股骨骨髓细胞染色体结构及数目异常细胞,观察雄性小鼠两侧附睾精子形态及各类畸形精子。结果表明:1)按5 000 mg/kg体重剂量经口灌胃,观察期内小鼠无中毒症状、大体剖检无异常变化、死亡率为0%。2)阳性对照组小鼠染色体畸变细胞率极显著高于阴性对照组(P<0.01),植物炭黑3个剂量组小鼠染色体畸变细胞率与阴性对照组差异不显著(P>0.05)。3)阳性对照组小鼠精子畸形率极显著高于阴性对照组(P<0.01),植物炭黑3个剂量组小鼠精子畸形率与阴性对照组差异不显著(P>0.05)。植物炭黑对小鼠经口急性毒性试验的LD50大于5 000 mg/kg体重,植物炭黑在1 250 mg/kg~5 000mg/kg体重剂量经口染毒后,小鼠骨髓细胞染色体畸变试验结果及小鼠精子畸形试验结果均为阴性。
【Abstract】 This experiment was conducted to evaluate the toxicological safety of plant carbon. The acute toxicity test of mice(LD50 determination) was performed by oral gavage at a dose of 5 000 mg/kg.BW of plant carbon. The poisoning symptoms, gross anatomy and death of mice were observed during the test period. Mouse bone marrow cell chromosome aberration test and mouse sperm malformation test were performed with cyclophosphamide 40 mg/kg.BW as positive control group and 1% sodium carboxymethyl cellulose as negative control group. Plant carbon was set up in three dose groups of 5 000 mg/kg.BW, 2 500 mg/kg.BW and 1 250 mg/kg.BW. All medicine were orally administered to observe the chromosomal structure and number of abnormal cells in the femur bone marrow cells of the test mice, and to observe the bilateral epididymal sperm morphology and various types of abnormal sperm of the male mice. The results showed as follows: 1) The mice were fed with 5 000 mg/kg.BW by oral gavage. During the observation period, the mice had no toxic symptoms, no abnormal changes in gross anatomy, and the mortality rate was 0%. 2) The rate of chromosome aberration cells in the positive control group was significantly higher than that in the negative control group(P<0.01). There was no significant difference in the rate of chromosome aberration cells between the three dose groups of plant carbon and the negative control group(P>0.05). 3)The sperm deformity rate of the positive control group was significantly higher than that of the negative control group(P<0.01). There was no significant difference in sperm deformity rate between the three dose groups of plant carbon and the negative control group(P>0.05). The LD50 of plant carbon in acute oral toxicity test in mice was greater than 5 000 mg/kg.BW. After oral exposure of plant carbon in the dose range of 1 250 mg/kg.BW ~5 000 mg/kg.BW, the results of chromosome aberration test and sperm deformity test in mice were negative.
