阿魏酸姜黄素酯衍生物对蜡样芽孢杆菌感染叉尾鮰后细胞因子及抗氧化酶的影响
Effect of Curcumin Feruferate Derivatives on Cytokines and Antioxidant Enzymes of Channel Catfish Infected with Bacillus cereus
【摘要】 为增强阿魏酸的生物活性,确定阿魏酸姜黄素酯衍生物对致病菌蜡样芽孢杆菌的防治效果,以阿魏酸和姜黄素为原料,通过酯化反应合成阿魏酸姜黄素酯衍生物(FA-CUR),产物经~1H-NMR、HRMS(ESI)进行表征。60条叉尾鮰均分为3组,分别为试验组、模型对照组和阴性对照组,试验组为FA-CUR给药3 d后人工感染蜡样芽孢杆菌,模型对照组为人工感染蜡样芽孢杆菌组,分别于感染后第4天采集组织与血清,采用荧光定量PCR检测各组鱼组织中炎症因子mRNA的表达以及试剂盒测定血清中抗氧化指标,确定FA-CUR对致病菌蜡样芽孢杆菌的抑菌效果。结果发现,经表征确定FA-CUR为所需化合物,且其对DPPH自由基、ABTS~+自由基的清除率随浓度的升高而升高。叉尾鮰经FA-CUR预防给药后,试验组中鱼组织炎症因子(IL-4r、IL-6、caspase-3、caspase-8、TNF-α、IFN-γ、caspase-9、IL-10)mRNA表达水平均显著下降;模型对照组血清中T-AOC及SOD含量显著上升,表明鱼的抗蜡样芽孢杆菌感染能力得到了增强。结果表明FA-CUR具有较好的抗氧化与抗菌活性,具有开发成新药物的潜力。
【Abstract】 To enhance the biological activity of ferulic acid, the control efficacy of curcumin ester derivatives of ferulic acid against the pathogenic bacteria B.cereus was determined.The ferulic acid derivative(FA-CUR) was synthesized with ferulic acid and curcumin, and the products were characterized by ~1H-NMR and HRMS(ESI).The 60 channel catfish were divided into 3 groups: test group, model control group and negative control group.The channel catfish in test group was artificially infected with B.cereus after FA-CUR administration for 3 days; The channel catfish in model control group were artificially infected with B.cereus.Then, the tissue and serum were collected on the fourth day.The mRNA expressions of inflammation factors in fish tissue was determined using quantitative PCR.The antioxidant enzyme activity in serum was also determined.The results indicated that the esterification process of FA-CUR was effective, and its removing activity of DPPH radicals and ABTS~+ radicals increased with increasing concentrations.After treatment with FA-CUR,the mRNA expressions of tissue inflammatory factors(IL-4r, IL-6,caspase-3,caspase-8,TNF-α,IFN-γ,caspase-9,IL-10) decreased significantly in test group.The T-AOC and SOD in model control group increased significantly, indicating that the resistant ability against B.cereus infection enhanced.Studies showed that FA-CUR has good antioxidant and antibacterial activities, and it has the potential to develop into new feed additives.
