高效液相色谱-荧光检测法测定分散黄82的含量
Determination of Disperse Yellow 82 Content by High Performance Liquid Chromatography-Fluorescence Detection
【摘要】 建立利用高效液相色谱-荧光检测法测定分散黄82含量的方法。色谱柱为WatersSunFireTM C18(5μm 4.6×250 mm),荧光检测器的激发波长为425 nm,发射波长495 nm,流动相为乙腈-四氢呋喃-水(体积比50∶20∶30),流速为0.8 mL/min,柱温30℃,进样体积10μL。分散黄82的进样质量浓度在30~3 000μg/L时,峰面积与进样浓度呈良好的线性关系,相关系数为r=0.999 5,回收率为98.26%~100.23%,回收率测定结果的相对标准偏差为0.82%(n=5)。
【Abstract】 A method to determine Disperse Yellow 82 content by high performance liquid chromatography with fluorescence detection was established. Chromatographic column: Waters SunFireTM C18(5 μm 4.6×250 mm); the excitation wavelength of the fluorescence detector was 425 nm and the emission wavelength was 495 nm; the mobile phase was acetonitrile-tetrahydrofuran-water( volume ratio 50∶20∶30); the flow rate was 0.8 mL/min; the column temperature was 30 ℃; and the injection volume was 10 μL. When the injection concentration of Disperse Yellow 82 was 30-3 000 μg/L, there was a good linear relationship between the peak area and the concentration of the sample, the correlation coefficient was 0.999 5, the recovery rate was 98.26 %-100.23 %, and the RSD of the recovery rate was 0.82%(n=5).
