miR-19a对民猪前脂肪细胞分化影响的研究
Effects of miR-19a on differentiation of preadipocytes in Min pigs
【摘要】 为探究miR-19a对民猪脂肪细胞分化过程的调控机制,研究采用双荧光素酶报告载体、实时荧光定量PCR构建组织表达谱、过表达和抑制miR-19a后对成脂标志基因和脂肪酸代谢标志基因的影响,油红O染色等生物学技术分析miR-19a对前脂肪细胞成脂的影响。结果表明,miR-19a与KLF13呈靶向关系,过表达miR-19a抑制成脂标志基因表达,促进脂肪酸代谢标志基因表达,抑制脂滴形成。抑制miR-19a可促进成脂标志基因表达,抑制脂肪酸代谢标志基因表达,促进脂滴形成。综上,试验验证miR-19a可抑制前脂肪细胞成脂分化,确定KLF13为miR-19a靶基因,为进一步研究microRNA对民猪前脂肪细胞分化的调控机制提供参考。
【Abstract】 The aim was to investigate the effect of miR-19a on the differentiation process of adipocytes in Min pigs and to provide a basis for further researches on the molecular mechanism of adipocyte differentiation in Min pigs. The effects of miR-19a on preadipocyte lipogenesis were analyzed by biological techniques such as dual luciferase reporter vector, real-time fluorescence quantitative PCR to construct tissue expression profiles, detection of the effects on lipogenic marker genes and fatty acid metabolism marker genes after overexpression and inhibition of miR-19a, and oil red O staining. The results showed that miR-19a and KLF13 were targeting relationship. Overexpression of miR-19a inhibited lipogenesis marker gene expression, promoted fatty acid metabolism marker gene expression and inhibited lipid droplet formation; inhibition of mi R-19a promoted lipogenesis marker gene expression,inhibited fatty acid metabolism marker gene expression and promoted lipid droplet formation. In summary, this experiment verified that miR-19a could inhibit preadipocyte lipogenic differentiation and identified KLF13 as the target gene of miR-19a. It also provided a reference for further study on the regulation of microRNA on preadipocyte differentiation in civilian pigs.
