玉米CMS-S型不育系花药的转录组分析

【摘要】 为探索玉米CMS-S型雄性不育的分子机制,以不育系郑58cms-Q1261和保持系郑58单核晚期的花药为材料进行转录组分析,发现14 565个差异表达基因(DEGs),其中7 551个上调表达,7 014个下调表达。GO富集分析显示这些DEGs主要富集在代谢过程、细胞和催化活性等方面。KEGG显著富集的主要代谢通路为碳代谢、淀粉和蔗糖代谢、氧化磷酸化、糖酵解/糖异生、氨基糖和核苷酸糖代谢等。选取12个DEGs进行qRT-PCR分析,结果与RNA-Seq数据一致,证明转录组数据的准确性。进一步对DEGs分析,发现2个果胶裂解酶超家族蛋白、5个果胶裂解酶类,3个热休克蛋白和2个bHLH转录因子与雄性不育有关。

【Abstract】 To explore the molecular mechanism of CMS-S type male sterility in maize, transcriptome analysis of anthers was conducted at the late mononuclear stage using sterile line Zheng58cms-Q1261 and maintainer line Zheng58 at the late mononuclear stage. A total of 14 565 differentially expressed genes(DEGs) were identified including 7 551 up-regulated and 7 014 down-regulated genes. GO enrichment analysis showed that these DEGs were mainly enriched in metabolic processes, cellular components, and catalytic activity. KEGG pathway analysis revealed significant enrichment in metabolic pathways such as carbon metabolism, starch and sucrose metabolism,oxidative phosphorylation, glycolysis/gluconeogenesis, amino sugar and nucleotide sugar metabolism. 12 DEGs were selected for qRT-PCR analysis, and the results were consistent with RNA-seq data, demonstrating the accuracy of the transcriptome data. Further analysis of the DEGs revealed the male sterility related genes including 2 pectinase superfamilies, 5 pectinases, 3 heat shock proteins, and 2 bHLH transcription factors.
