基于潮湿环境下缺氧-线粒体损伤病理机制探究湿胜阳微理论本质
Exploration of the essence of Shisheng Yangwei theory based on pathological mechanism of hypoxia injury by external dampness
【摘要】 目的:基于潮湿环境下缺氧-线粒体损伤损伤病理机制探究湿胜阳微理论本质。方法:将22只雄性BALB/c小鼠随机分为对照组与潮湿组,每组11只。对照组饲养环境:温度23℃,相对湿度55%;潮湿组温度23℃,相对湿度85%。造模第10天各组随机选取5只小鼠,腹腔注射HypoxyprobeTM缺氧探针2 h后取材,透射电镜观察回肠组织线粒体变化;Western Bolt检测回肠与肝脏组织中缺氧诱导因子-1β(HIF-1β)与诱导型一氧化氮合酶(iNOS);免疫荧光法检测回肠组织中缺氧探针活化情况。造模第14天取材余下小鼠,免疫荧光检测回肠线粒体内膜蛋白Mic60;荧光定量PCR检测线粒体DNA;ELISA检测回肠组织中白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)。结果:与对照组比较,潮湿组小鼠回肠组织部分线粒体呈现缺氧相关结构受损,Mic60荧光面积占比显著减少(P<0.05),肝脏组织中HIF-1β含量显著升高(P<0.05),IL-6、TNF-α含量显著升高(P<0.01)。结论:潮湿环境下出现了缺氧诱导的细胞内线粒体损伤,导致机体出现相关症状;这似乎与湿胜阳微理论相通。
【Abstract】 Objective:To explore the essence of Shisheng Yangwei theory based on the pathological mechanism of hypoxic and mitochondrial damage in humid environment.Methods:A total of 22 male BALB/c mice were randomly divided into control group and wet group with 11 mice in each group.Control group feeding environment:temperature:23℃,relative humidity:55%;Humidity group temperature:23°C,relative humidity 85%.On the 10th day of modeling,5 mice were randomly selected from each group,and the samples were taken 2 hours after HypoxyprobeTMhypoxia probe was injected intraperitoneally,and the mitochondrial changes of ileum were observed by transmission electron microscopy.Hypoxia-inducible factor-1β (HIF-1β) and inducible nitric oxide synthase (iNOS) in ileum and liver were detected by Western Blot.The activation of hypoxic probe in ileum was detected by immunofluorescence.On the 14th day of modeling,the remaining mice were collected and the ileal mitochondrial intimal protein (Mic60) was detected by immunofluorescence.Mitochondrial DNA was detected by PCR.Interleukin-6 (IL-6) and tumor necrosis factor α (TNF-α) were detected by ELISA.Results:Compared with the control group,some mitochondria in the wet group showed hypoxia-related structure damage,Mic60 protein decreased (P<0.05),HIF-1βcontent increased (P<0.05),IL-6 and TNF-α contents significantly increased (P<0.01).Conclusion:Hypoxia-induced intracellular mitochondrial damage occurs in humid environment,leading to related symptoms.This seems to be related to Shisheng Yangwei theory.
