人参皂苷Rh2对糖尿病肾病变大鼠肾损伤的保护作用及其机制
Protective effect of ginsenoside Rh2 on kidney injury in rats with diabetic kidney lesions and its mechanism
【摘要】 目的:探讨人参皂苷Rh2对糖尿病肾病变(DKL)大鼠肾脏损伤的保护作用,并阐明其作用机制。方法:通过高脂高能量饲料配以链脲佐菌素(STZ)腹腔注射方法制备DKL大鼠模型。将30只SD成模大鼠随机分为模型组、低剂量(10 mg·kg-1) Rh2组和高剂量(20 mg·kg-1) Rh2组,每组10只,另选取10只SD大鼠作为正常对照组。检测各组大鼠体质量和肾脏质量,并计算肾指数,干预8周后全自动分析仪检测各组大鼠血清肌酐(Scr)、尿素氮(BUN)和24 h尿蛋白(UP)水平,酶联免疫吸附测定(ELISA)法检测各组大鼠肾组织中Ⅳ型胶原(ColⅣ)水平,Western blotting法检测各组大鼠肾组织中转化生长因子β1 (TGF-β1)、Smad3和Smad7蛋白表达水平。结果:与正常对照组比较,模型组大鼠体质量降低(P<0.01),肾脏质量和肾指数明显升高(P<0.01),血清中Scr、BUN和24 h UP水平均升高(P<0.01),肾组织中ColⅣ水平升高(P<0.01),肾组织中TGF-β1和Smad3蛋白表达水平升高(P<0.01),而肾组织中Smad7蛋白表达水平降低(P<0.01)。与模型组比较,低和高剂量Rh2组大鼠体质量升高(P<0.05或P<0.01),肾脏质量和肾指数均降低(P<0.05或P<0.01),血清中Scr、BUN和24 h UP水平均明显降低(P<0.05或P<0.01),肾组织中ColⅣ水平降低(P<0.01),肾组织中TGF-β1和Smad3蛋白表达水平降低(P<0.01),肾组织中Smad7蛋白表达水平升高(P<0.01)。结论:人参皂苷Rh2可抑制DKL大鼠肾脏纤维化,改善肾功能,对DKL大鼠肾脏有保护作用,其机制可能与Rh2调节TGF-β1/Smads信号通路表达有关。
【Abstract】 Objective:To discuss the protective effect of ginsenoside Rh2 on kidney injury in the rats with diabetic kidney lesions(DKL),and to clarify its mechanism. Methods:The DKL rat model was established by using the high-fat and high-energy diet combined with intraperitoneal injection of streptozotocin(STZ). Thirty SD model rats were randomly divided into model group, low dose(10 mg·kg-1) of Rh2 group and high dose(20 mg·kg-1) of Rh2 group, and there were 10 rats in each group;other ten SD rats were regared as normal control group. The body weights and kidney weights of the rats in various groups were detected,and the kidney index was caculated;after intervented for 8 weeks, automatic analyzer was used to detect the levels of serum creatinine(Scr), urea nitrogen(BUN) and 24 h urinary protein(UP)in serum of the rats in various groups;enzyme linked immunosorbent assay(ELISA) method was used to detect the levels of collagen type Ⅳ(Col Ⅳ) in kidney tissue of the rats in various groups; Western blotting method was used to detect the expression levels of transforming growth factor-β1(TGF-β1),Smad3 and Smad7 proteins in kidney tissue of the rats in various groups.Results:Compared with normal control group, the body weight of the rats in model group was decreased(P<0. 01),the kidney weight and kidney index were significantly increased(P<0. 01), the levels of Scr, BUN and 24 h UP in serum were increased(P<0. 01), the level of Col Ⅳ in kidney tissue was increased(P<0. 01), the expression levels of TGF-β1 and Smad3 proteins in kidney tissue were increased(P<0. 01), and the expression level of Smad7 protein in renal tissue was decreased(P<0. 01). Compared with model group,the body weight of the rats in low and high doses of Rh2 groups were inecreased(P<0. 05 or P<0. 01), the kidney weight and kidney index were decreased(P<0. 05 or P<0. 01),the levels of Scr, BUN and 24 h UP in serum were significantly decreased(P<0. 05 or P<0. 01), the level of Col Ⅳ in kidney tissue was decreased(P<0. 01), the expression levels of TGF-β1 and Smad3 proteins in kidney tissue were decreased(P<0. 01), and the expression level of Smad7 protein in kidney tissue was increased(P<0. 01). Conclusion:Ginsenoside Rh2 can inhibit the kidney fibrosis and improves the kidney function in the DKL rats, and has a protective effect on the kidney of DKL rats; its mechanism may be related to regulating the expression of TGF-β1/Smads signaling pathway.
