兰州百合组培快繁技术研究

【摘要】 本试验以兰州百合带腋芽的嫩茎为外植体,通过无菌芽诱导、丛生芽增殖及芽苗生根培养,对兰州百合进行组织培养及快速繁殖技术研究。结果表明:选用6-BA和NAA两种不同浓度的植物生长调节剂对不定芽进行诱导培养,筛选出诱导不定芽的适宜培养基为MS+6-BA 1.0 mg/L+NAA 0.5 mg/L,分化率可达88.00%,平均苗高为3.10 cm;选用6-BA和IBA两种不同浓度的植物生长调节剂进行继代增殖培养时,筛选出丛生芽增殖的适宜培养基为MS+IBA 0.3 mg/L+6-BA 0.5 mg/L,扩繁系数可达3.40,平均苗高4.60 cm;选用IAA和IBA两种不同浓度的植物生长调节剂进行生根培养,筛选出生根培养的适宜培养基为1/2MS+IAA 0.1 mg/L+IBA 0.2 mg/L,生根率90.00%,平均根数3.20条。

【Abstract】 In this paper,the tender stem with axillary buds of Lanzhou lily was used as explant,and the tissue culture and rapid propagation technology of Lanzhou lily were studied by inducing aseptic buds,multiplying clustered buds and rooting of buds.The results showed that he adventitious buds were induced and cultured with 6-BA and NAA at different concentrations,and the optimal medium for inducing adventitious buds was MS+6-BA 1.0 mg/L+NAA0.5 mg/L,the differentiation rate was 88.00%,and the average seedling height was 3.10 cm.When two plant growth regulators (6-BA and IBA) with different concentrations were selected for subculture,the optimal medium for the multiplication of clustered buds was MS+IBA 0.3 mg/L+6-BA 0.5 mg/L,the multiplication times could reach 3.40,and the average seedling height was 4.60 cm.Two plant growth regulators (IAA and IBA) with different concentrations were used for rooting culture,the suitable medium for rooting culture was 1/2MS+IAA 0.1 mg/L+IBA 0.2 mg/L,the rooting rate was 90.00%,and the average number of roots was 3.20.
