基于高通量和传统培养法分析白首乌根际真菌多样性

【摘要】 探讨白首乌根际土壤真菌群落组成结构与分布规律,为优选根际促生菌、提高仿野生种植技术提供科学理论依据。综合Illumina MiSeq高通量测序技术和传统分离培养技术,分析测定3个产地2个根茎深度的白首乌根际土壤真菌群落的丰富度和多样性,分离鉴定可培养根际真菌的种类。6个样品共得到真菌1 589个OTUs,分属于11门35纲84目142科237属,子囊菌门(Ascomycota)和被孢霉门(Mortierellomycota)是样品共同的优势菌门;6个样品通过传统培养法共分离得到103个形态型,分为3门8纲10目12科16属37种,优势菌属为木霉属(Trichoderma)和镶刀菌属(Fusarium)。综合比较,高通量测序技术可以得到更全面的菌落数据,传统分离培养技术可以纯化出适应性强的菌株;不同产地和根茎深度样品间土壤真菌群落存在显著差异性。

【Abstract】 The composition, structure and distribution of rhizosphere fungal community of Cynanchum bungei Decne were discussed so as to provide a scientific theoretical basis for optimizing rhizosphere growth promoting fungi and improving imitating wild planting technology. By integrating Illumina MiSeq High-throughput sequencing technology and traditional isolation and culture technology, the richness and diversity of fungal communities in the rhizosphere soil of Cynanchum bungei Decne at two rhizome depths from three habitats were analyzed and determined, and the cultivable rhizosphere fungi were isolated and identified. A total of 1589 OTUs were obtained from 6 samples, belonging to 11 phyla, 35classes, 84 orders, 142 families and 237 genera. Ascomycota and Mortierellomycota are the common dominant phyla of the samples. A total of 103 morphotypes were isolated from 6 samples by traditional culture method, which were divided into 3 phyla, 8 classes, 10 orders, 12 families, 16 genera and 37 species. The dominant genera are Trichoderma and Fusarium. By comprehensive comparison, high-throughput sequencing technology can obtain more comprehensive colony data, and traditional isolation and culture technology can purify strains with strong adaptability. There were significant differences in soil fungal communities among samples from different producing areas and rhizome depths.
