不同浓度激素配比对防风愈伤组织诱导及再分化的影响

【摘要】 以防风的有活力的种子为材料,通过栽种土培苗,比较不同来源外植体愈伤组织的形成能力,并通过不同激素配比调整,研究愈伤组织分化生根、生芽的适宜条件,进而建立防风组培苗再生体系。结果表明,以幼叶为外植体所诱导出的愈伤组织分化情况最佳,最佳诱导培养基为MS+1.5 mg/L 2,4-D+0.5 mg/L 6-BA,诱导率达86.6%;最佳继代培养基为MS+1.0 mg/L 6-BA+0.6 mg/L NAA,增殖倍数为3.2;最佳诱导丛生芽培养基为MS+0.3 mg/L 6-BA,生芽率为100%;最佳诱导不定根最佳培养基为MS+0.2 mg/L 6-BA+0.4 mg/L IAA,生根率为46.8%。

【Abstract】 Using the active seeds of Saposhnikovia divaricata as materials, the ability of callus formation from explants from different sources was compared by planting soil culture seedlings, and through adjusting the ratio of different hormones, the suitable conditions for callus differentiation, rooting and budding were studied, and then the regeneration system of Saposhnikovia divaricata tissue culture seedlings was established.The results showed that the callus differentiation induced from young leaves was the best, and the optimal induction medium was MS+1.5 mg/L 2,4-D+0.5 mg/L 6-BA,with an induction rate of 86.6%.The optimal subculture medium was MS+1.0 mg/L 6-BA+0.6 mg/L NAA,the proliferation rate was 3.2.The optimal medium for inducing clustered buds was MS+0.3 mg/L 6-BA,and the budding rate was 100%;The optimal medium for inducing adventitious roots was MS+0.2 mg/L 6-BA+0.4 mg/LIAA,and the rooting rate was 46.8%.
