不同培养条件对长枝木霉SMF2和哈茨木霉T39生长与产孢的影响

【摘要】 采用十字交叉法和血球计数法分析不同培养基、温度、光照、酸碱度、碳源和氮源等对长枝木霉SMF2和哈茨木霉T39菌丝生长和孢子形成的影响,明确两种木霉生长繁殖的最佳条件,指导其人工扩繁和工厂化生产。结果表明,SMF2和T39在含碳源和有机氮的PDA、CDA培养基中,常温、黑暗、非强酸强碱培养条件下其菌丝生长良好,光照、果糖、牛肉膏培养条件更有利于二者孢子形成;温度、酸碱度和培养基是影响SMF2和T39孢子形成的主要因素,35℃、pH值为7、PDA培养基最利于SMF2产孢,30℃、pH值为6、CDA培养基最利于T39产孢;相同条件下,SMF2的菌丝生长略快,而T39的产孢能力更强;单糖和有机氮更有利于促进SMF2和T39产孢。

【Abstract】 The effects of different media, temperature, light, pH, carbon sources and nitrogen sources on the hypha growth and spore formation of Trichoderma longibrachiatum SMF2 and Trichoderma harzianum T39 were analyzed by cross method and blood cell counting method. It was aimed to clarify the optimal conditions for the growth and reproduction of the two Trichoderma species, and to guide their artificial propagation and factory production. The results showed that SMF2 and T39 had better hypha growth under room temperature, dark, non-strong acid and alkali culture conditions in PDA and CDA media containing carbon source and organic nitrogen, and the culture conditions of light, fructose and beef paste were more conducive to the spore formation. Temperature, pH and medium were the main factors affecting the formation of SMF2 and T39 spores; 35 ℃, pH=7 and PDA medium were the most conducive to SMF2 spore production, and 30 ℃, pH=6 and CDA medium were the most conducive to T39 spore production. Under the same conditions, the hypha growth rate of SMF2 was slightly faster, and the spore production capacity of T39 was stronger. Monosaccharides and organic nitrogen were more conducive to SMF2 and T39 sporulation.
