槲蕨孢子育苗关键技术研究
Key Technology of Spore Breeding for Drynaria fortune (Kunze)
【摘要】 目的:为槲蕨孢子育苗提供技术支持。方法:以不同基质进行直播,观测其萌发率和生长势;在原叶体快速生长期以1/2MS、1/4MS、1/8MS营养液、清水定期喷施,研究原叶体受精率。结果:不同基质中槲蕨孢子萌发率呈显著差异,其中以“细腐殖土”(过3号筛)“、中号腐殖土”(过1号筛)“、细河沙”(过3号筛)、中号河沙(过1号筛)、紫色细壤土(过3号筛)、紫色中壤土(过1号筛)为基质,孢子萌发率高于82.0%,原叶体长势好;在原叶体快速生长期,环境湿度90%有利于原叶体受精及孢子苗生长;以1/4MS营养液间隔3 d喷1次,原叶体生长良好,受精率最高,为41.00%。结论:基质对槲蕨孢子萌发和生长有影响,原叶体生长期喷施营养液能促进其生长和受精。
【Abstract】 Objective:To support for the spore breeding of Drynaria fortune,using different substrates for live broadcast to observe the germination rate and growth vigor;During the rapid growth period of the prothallium,1/2MS,1/4MS,1/8MS nutrient solution and water were sprayed regularly for management to study the fertilization rate.Results:Different substrates showed significant differences in the germination rate of Quercus fern spores. Among them, "humus soil"(passing No. 3 sieve), "humus soil"(passing No. 1 sieve),and "river sand"(passing 3 No. sieve), "river sand"(passing the No. 1 sieve), "purple fine loam"(passing the No. 3 sieve),and "purple medium loam"(passing the No. 1 sieve)were used as substrates. The spore germination rate was higher than other substrates,and the prothallium grows well.In the rapid growth period of prothallium,90% environmental humidity was conducive to the fertilization of prothallium and the growth of spore seedlings;sprayed with 1/4MS nutrient solution every 3 d,the prothallium grew well,and the fertilization rate of prothallium was the highest,41.00%.Conclusion:The substrate has an effect on the germination and growth of Drynaria fortune fern spores. Spraying nutrient solution during the growth period of the prothallium can promote its growth and fertilization.
