云南民族药理肺散防治急性肺损伤的作用及网络药理学分析

【摘要】 目的:考察云南民族药理肺散防治急性肺损伤(ALI)的作用及机制。方法:复制小鼠气管滴注脂多糖(LPS)致ALI模型,以及尾静脉注射花生四烯酸(AA)诱导小鼠急性肺栓塞模型,考察理肺散的作用。采用网络药理学方法,利用SwissADME与SwissTargetPrediction分析平台,筛选理肺散的化学成分及对应靶点,采用GeneCards、OMIM和TTD数据库检索ALI疾病相关靶点,构建“药物-成分-疾病-靶点”网络,进行信号通路富集分析。结果:理肺散对AA诱导的急性肺栓塞有明显的保护作用,可显著提高肺栓塞小鼠的存活率(P<0.01)。小鼠气管滴注LPS后,肺组织湿/干质量比值(W/D)、肺泡灌洗液中肿瘤坏死因子α(TNF-α)和髓过氧化物酶(MPO)等炎症因子含量显著升高,超氧化物歧化酶(SOD)水平显著下降(P<0.01);给予高剂量理肺散预处理后,可明显降低肺组织W/D值,减轻肺水肿,降低肺泡灌洗液中TNF-α、MPO的含量,并提高SOD的水平(P<0.05或P<0.01)。“药物-成分-疾病-靶点”网络及基因功能注释结果显示,理肺散对ALI的防治作用与调控炎症、氧化应激、血小板激活等通路相关。结论:理肺散可能通过调控炎症等信号通路发挥防治急性肺损伤的作用。

【Abstract】 Objective: To investigate the effects and mechanism of Yunnan ethnic herb Lifeisan in the prevention and treatment of acute lung injury(ALI).Methods: ALI model induced by lipopolysaccharide(LPS) intratracheal instillation and pulmonary embolism model induced by arachidonic acid(AA) intravenous injection in mice were used to verify the roles of Lifeisan. The method of network pharmacology was used to analyze the mechanism of Lifeisan. The active components and related targets of Lifeisan were screened from SwissADME and SwissTargetPrediction analysis platform, the disease related targets of ALI were searched by using GeneCards, OMIM and TTD database. And then “herb-components-disease-target” network was constructed, and signal pathway enrichment analysis was carried out. Results: Lifeisan showed obvious protective effect on pulmonary embolism induced by AA, and improved the survival rate of mice significantly(P<0.01). After intratracheal instillation of LPS in mice, the lung tissue wet/dry weight ratio(W/D), and the contents of TNF-α and MPO in alveolar lavage fluid increased significantly, while the level of SOD decreased significantly(P<0.01). After pretreatment with high dose of Lifeisan, the W/D, and the contents of TNF-α and MPO decreased significantly, while the level of SOD increased significantly(P<0.05 or P<0.01). The “herb-components-disease-target” network and Gene function annotation showed that the effects of Lifeisan on ALI were related to the regulation of inflammation, oxidative stress and platelet activation.Conclusion: Lifeisan may play a role in the treatment of acute lung injury by regulating signaling pathways such as inflammation.
