益气化浊胶囊预防高脂饮食大鼠胰岛素抵抗的作用初探

【摘要】 目的:观察益气化浊胶囊对高脂饮食大鼠胰岛素抵抗(Insulin Resistance,IR)的预防作用,并初步探讨其可能的作用机制。方法:将SD大鼠随机分为空白对照组、模型对照组、盐酸二甲双胍组(0.2 g·kg-1·d-1)和益气化浊胶囊高、中、低剂量组(2 g·kg-1·d-1、1 g·kg-1·d-1、0.5 g·kg-1·d-1),每组10只。灌胃给药,每日1次,连续8周。给药的同时造模,造模采用高脂饮食喂养诱导IR法。实验期间观察大鼠一般情况,灌胃第8周末,测量体质量、身长,计算Lee’s指数,快速血糖仪测空腹血糖,酶联免疫吸附法检测血清白脂素、胰岛素水平,计算稳态模式评估法胰岛素抵抗指数(Homeostasis Model Assessment of Insulin Resistance Index,HOMA-IR)来评价IR水平。结果:灌胃第8周末时,模型对照组体质量、身长、Lee’s指数、胰岛素水平、HOMA-IR、白脂素水平显著高于空白对照组(P <0.01);而二甲双胍组、益气化浊胶囊高剂量组体质量、身长显著低于模型对照组(P <0.01或P <0.05),二甲双胍组Lee’s指数显著低于模型对照组(P <0.05),二甲双胍组及益气化浊胶囊高、中剂量组胰岛素水平、HOMA-IR、白脂素水平显著低于模型对照组(P <0.01或P <0.05)。结论:益气化浊胶囊可在一定程度上预防高脂饮食大鼠IR,其作用机制可能与血清白脂素水平的降低有关。

【Abstract】 Objective: To observe the preventive effect of the Yiqi Huazhuo capsules( 益气化浊胶囊) on insulin resistance in rats with high-fat diet, and to explore its possible mechanism. Methods: SD rats were randomly divided into the blank control group, the model control group, the metformin hydrochloride group(0.2 g·kg-1·d-1) and the high, middle and low dose groups of the Yiqi Huazhuo capsules(2 g·kg-1·d-1, 1 g·kg-1·d-1, 0.5 g·kg-1·d-1), 10 rats in each group. Gavage was given once a day, for 8 consecutive weeks. At the same time of administration, the IR rats models were made by high-fat diet feeding induction method. Results: At the end of the 8th week, the body weight, body length, Lee’s index, insulin level, HOMA-IR and asprosin level in the model control group were significantly higher than those in the blank control group(P<0.01), while the body weight, and body length in the metformin hydrochloride group and the high dose group of the Yiqi Huazhuo capsules were significantly lower than the model control group(P<0.01 or P<0.05), the Lee’s index in the metformin hydrochloride group was significantly lower than the model control group(P<0.05), and the insulin level, HOMA-IR, and asprosin level in the metformin hydrochloride group, the high and middle dose groups of the Yiqi Huazhuo capsules were significantly lower than the model control group(P<0.01 or P<0.05). Conclusion: The Yiqi Huazhuo capsules can prevent from IR in rats with high-fat diet to some extent, and its mechanism may be related to the decrease of serum asprosin level.
