脑心通胶囊对大鼠心梗后心室重构的影响研究
Effect of Naoxintong Capsule on Ventricular Remodeling after Myocardial Infarction in Rats
【摘要】 目的 观察脑心通胶囊对大鼠心肌梗死后心室重构的影响。方法 将40只大鼠随机分为假手术组、模型组、脑心通组和他汀组,采用冠状动脉左前降支结扎法构建心肌梗死后心室重构大鼠模型(1周),模型构建成功后分别给予生理盐水、脑心通胶囊和阿托伐他汀钙片灌胃4周。在实验第5周采用Doppler超声检测大鼠心功能,HE染色和Masson染色观察心肌病理变化,QPCR检测心肌LXRα、NOX、ROS、COLΙ、COLⅢmRNA的表达。结果 与假手术组比较,模型组大鼠正常心肌减少、心肌纤维化,左室舒张末期内径(LVIDd)、左室收缩末期内径(LVIDs)升高(P <0.05),左室射血分数(LVEF)、短轴收缩率(FS)、每搏输出量(SV)、排血量(CO)降低(P <0.05),LXRα mRNA表达降低(P <0.05),NOX、COLⅠ、COLⅢm RNA表达增加(P <0.05),ROS活性增加(P <0.05);与模型组比较,脑心通组大鼠正常心肌纤维化减轻,LVIDd、LVIDs降低(P <0.05),EF、FS、SV、CO升高(P <0.05),LXRα mRNA表达升高(P <0.05),NOX、COLΙ、COLⅢmRNA表达降低(P <0.05),ROS活性降低(P <0.05)。结论 脑心通胶囊可能通过上调LXRα m RNA,抑制NOX、COLΙ、COLⅢmRNA的表达和ROS活性,从而延缓心肌梗死后心室重构。
【Abstract】 Objective:To investigate the effect of Naoxintong Capsules on ventricular remodeling after myocardial infarction in rats. Methods:A total of 40 rats were randomly divided into the sham operation group,model group,Naoxintong group and statin group. The left anterior descending coronary artery ligation method was used to construct a rat model of ventricular remodeling after myocardial infarction(1 week). After successful model construction,they were given normal saline,Naoxintong Capsules and atorvastatin calcium tablets by gavage for 4weeks. In the 5th week,Doppler ultrasound was used to detect the cardiac function of rats;HE staining and Masson staining were used to observe the myocardial pathological changes,and QPCR was used to detect the expression of myocardial LXRα,NOX,ROS,COLⅠ,and COLⅢ mRNA. Results:Compared with the sham operation group,the model group had decreased normal myocardium,myocardial fibrosis,increased LVIDd and LVIDs(P <0.05),decreased LVEF,FS,SV,CO(P < 0.05),decreased LXRα m RNA expression(P < 0.05),increased NOX,COLⅠ,COLⅢ m RNA expression(P < 0.05),and increased ROS activity(P < 0.05);compared with the model group,Naoxintong group had decreased myocardial fibrosis,decreased LVIDd LVIDs(P < 0.05),increased EF,FS,SV,CO(P < 0.05),increased LXRα mRNA expression(P < 0.05),decreased NOX,COLΙ,COLⅢ m RNA expression(P <0.05),and decreased ROS activity(P < 0.05). Conclusion:Naoxintong Capsules may up-regulate LXRα mRNA,inhibit NOX,COLⅠ,COLⅢ mRNA expression and ROS activity,thereby delaying ventricular remodeling after myocardial infarction.
