积雪草酸对脑梗死大鼠神经保护作用研究
Protective effect of asiatic acid on the neurological function of rats with ischemic stroke
【摘要】 目的 探讨积雪草酸对缺血性脑卒中(IS)大鼠神功功能的保护作用及其机制。方法 75只健康、雄性清洁级Wistar大鼠随机分为假手术组,脑缺血组,积雪草酸-L组(50 mg/kg),积雪草酸-M组(100 mg/kg),积雪草酸-H组(200 mg/kg)。各组通过大脑中动脉线栓阻塞法构建缺血性脑卒中模型后给予积雪草酸治疗。3 d后评估大鼠神经功能。循序处死大鼠,取脑组织开展梗死体积、脑水肿、血脑屏障完整性检测,免疫荧光染色与Western blot试验。结果 与脑缺血组相比,积雪草酸可明显降低缺血性脑卒中大鼠神经功能评分(P<0.05),减少伊文思蓝穿透量(P<0.05)、脑水肿程度(P<0.05)、脑梗死体积(P<0.05)。积雪草酸可增加大鼠缺血区CD31/Ki67双阳性染色(P<0.05),且脑组织中VEGFA与Ang1蛋白表达水平上调(P<0.05),Claudin-5与ZO-1表达增加(P<0.05)。结论 积雪草酸对缺血性脑卒中具有治疗作用,其机制可能是提高VEGFA与Ang1表达,促进血管内皮细胞增殖,帮助血脑屏障结构的修复。
【Abstract】 Objective To investigate the protective effect and mechanism of asiatic acid on the neurological function of rats with ischemic stroke (IS).Methods A total of 75 healthy male clean-grade Wistar rats were randomly divided into a sham operation group,a cerebral ischemia group,an asiatic acid-L group (50 mg/kg),an asiatic acid-M group (100 mg/kg),and an asiatic acid-H group (200 mg/kg).The rats were treated with asiatic acid after the ischemic stroke model was established by the middle cerebral artery occlusion method.The neurological function of rats was evaluated 3 days later.The rats were killed in sequence,and the brain tissues were taken for the infarct volume test,brain edema test,blood-brain barrier integrity evaluation,immunofluorescence staining and Western blot test.Results Compared with the cerebral ischemia group,asiatic acid significantly reduced the neurological function score (P<0.05),the penetration of Evans blue (P<0.05) and the cerebral edema (P<0.05),the cerebral infarction volume (P<0.05) in rats with ischemic stroke.Asiatic acid increased the CD31/ Ki67 double positive staining in the ischemic area of rats (P<0.05).In addition,the expression levels of VEGFA and Ang1 in brain tissues were up-regulated (P<0.05),and the expressions of Claudin-5 and ZO-1 were increased (P<0.05).Conclusion Asiatic acid has a therapeutic effect on ischemic stroke,and its mechanism may be due to increasing the expression of VEGFA and Ang1,promoting the proliferation of vascular endothelial cells,and helping repair the blood-brain barrier structure.
