LncRNA SNHG12在恶性脑胶质瘤中的表达水平及临床意义
The expression levels and clinical significance of LncRNA SNHG12 in glioma
【摘要】 目的 基于生物信息学与临床样本相结合分析检测LncRNA SNHG12在恶性脑胶质瘤患者中的表达情况并探讨其与患者临床病理特征和预后的关系。方法 从GEO数据库和TCGA数据库中分析LncRNA SNHG12的表达水平以及其对恶性脑胶质瘤患者预后的影响。收集湘雅医院Ⅰ-Ⅳ级恶性脑胶质瘤患者和脑外伤患者或癫痫患者,采用实时荧光定量PCR(qRT-PCR)法检测LncRNA SNHG12在Ⅰ-Ⅳ级恶性脑胶质瘤组织以及正常脑组织中的表达,并对患者进行随访,分析LncRNA SNHG12表达对恶性脑胶质瘤预后的影响。结果 在恶性脑胶质瘤组织中LncRNA SNHG12表达水平显著高于正常脑组织,在高级别病理分级的恶性脑胶质瘤组织中的表达水平显著高于低级别(P<0.05);且LncRNA SNHG12的表达情况与患者年龄、性别以及病灶位置无关,与患者的肿瘤病理分级有关(Ⅰ-Ⅱ级vs.Ⅲ-Ⅳ级,P<0.0001);高表达LncRNA SNHG12的恶性脑胶质瘤患者的总存活率均显著低于低表达患者;同时,Cox模型多因素分析结果显示,患者的肿瘤病理分级是影响预后的独立危险因素。结论 基于生物信息学与临床样本相结合分析可知恶性脑胶质瘤患者中的LncRNA SNHG12表达上调,其表达水平与患者的肿瘤病理分级有关,可能成为恶性脑胶质瘤患者不良预后的生物标志物。
【Abstract】 Objective To explore the expression of long non-coding RNA SNHG12in glioma and the relationship with clinicopathological characteristics based on the combinative analysis of bioinformatics and clinical samples.Methods The expression level of LncRNA SNHG12and its influence on the prognosis of patients with malignant glioma were analyzed from GEO database and TCGA database.The glioma tissues were acquired from tumor patients who only underwent surgical resection at Xiangya Hospital(Changsha,Hunan China).Non-tumor specimen were obtained from patients with brain injured.The qRT-PCR was used to measure the expression levels of long non-coding RNA SNHG12in glioma tissues and non-tumor tissues.The relationship between long non-coding RNA SNHG12and clinicopathological features of glioma was analyzed.Moreover,all the patients were followed up to analyze the effects of the expressions of long non-coding RNA SNHG12on the prognosis.Results The expression level of LncRNA SNHG12in malignant glioma tissues was significantly higher than that in normal brain tissues,and the expression level of LncRNA SNHG12in malignant glioma tissues with high pathological grade was significantly higher than that of low-grade malignant glioma tissues(P<0.05).Moreover,the expression of LncRNA SNHG12was not related to the age,gender and lesion location of the patients,but was related to the pathological grade of the patients(GradeⅠ-Ⅱvs.GradeⅢ-Ⅳ,P<0.0001).The overall survival rate of malignant glioma patients with high expression of LncRNA SNHG12was significantly lower than that of patients with low expression.Cox model multivariate analysis results showed that the pathological grade of the patients could be considered as an independent poor prognostic indicators in gliomas.Conclusion Based on the combinative analysis of bioinformatics and clinical samples,the expression levels of LncRNA SNHG12are up-regulated in glioma tissues which are related to clinicopathological features and may be regarded as the biomarker of poor prognostic indicators in glioma patients.
