COL3A1在胃癌中的表达及意义
Expression and significance of COL3A1 in gastric cancer
【摘要】 目的检测胃癌及其正常组织中COL3A1的mRNA及蛋白表达水平,初步探究COL3A1与胃癌临床及病理特征的关系。方法从胃癌手术患者癌组织及其正常组织中收集新鲜标本各10例,石蜡标本各50例,分别使用实时定量PCR法(Quantitative Real-time PCR)和免疫组化法检测COL3A1mRNA及蛋白水平的表达。同时,收集受检患者临床资料,分析COL3A1的表达与胃癌临床病理特征(性别、年龄、分化程度、淋巴转移、TNM分期、肿瘤浸润深度及肿瘤大小)的关系。结果 (1)COL3A1在胃癌组织中mRNA水平表达量为6.86±3.07,其正常组织中COL3A1表达量仅2.48±1.25,差异有统计学意义(t=4.181,P=0.0013);对比在胃癌组织及其正常组织中COL3A1表达的蛋白水平,前者明显高于后者,差异有统计学意义(χ~2=10.256,P=0.001)。(2)COL3A1蛋白表达水平与TNM分期(P<0.001)、浸润深度(P=0.012)有关,而与性别(P=0.529)、年龄(P=0.880)、分化程度(P=0.869)、肿瘤大小(P=0.145)、淋巴结转移(P=0.416)等因素无关。结论 (1)COL3A1在胃癌组织中mRNA及蛋白水平的表达均显著高于正常组织;(2)COL3A1高表达与肿瘤分期及浸润深度有关,与性别、年龄、分化程度、肿瘤大小、淋巴结转移等无关。
【Abstract】 Objective To detect the mRNA and protein expression levels of COL3A1 in gastric cancer and its normal tissues,and to preliminarily explore the relationship between COL3A1 and the clinicopathological features of gastric cancer Methods Ten fresh samples and 50 paraffin samples were collected from normal tissues and cancer tissues of gastric cancer patients.Quantitative real-time PCR and immunohistochemistry were used to detect the expression of COL3A1 mRNA and protein levels,respectively At the same time,clinical data of patients were collected to analyze the relationship between COL3A1 expression and clinicopathological characteristics of gastric cancer(gender,age,degree of differentiation,lymphatic metastasis,TNM stage,depth of tumor invasion and tumor size).Results (1)The mRNA level of COL3A1 in gastric cancer tissues was 6.86±3.07,while that in normal tissues was only 2.48±1.25,the difference was statistically significant(t=4.181,P=0.0013).The protein level of COL3A1 in gastric cancer tissues and its normal tissues was significantly higher than that in gastric cancer tissues,and the difference was statistically significant(χ~2=10.256,P=0.001).(2)The expression level of COL3A1 protein was correlated with TNM stage(P<0.001)and depth of invasion(P=0.012),but not with gender(P=0.529),age(P=0.880),degree of differentiation(P=0.869),tumor size(P=0.145)and lymph node metastasis(P=0.416).Conclusion (1)The mRNA and protein levels of COL3A1 in gastric cancer tissues were significantly higher than those in normal tissues;(2)The high expression of COL3A1 was correlated with tumor stage and invasion depth,but not with gender,age,degree of differentiation,tumor size,lymph node metastasis.
