管花肉苁蓉醇提物抗小鼠运动性疲劳的效果

【摘要】 为了探讨管花肉苁蓉(CtW)抗小鼠运动性疲劳的效果,本试验将60只健康雄性昆明小鼠分为5个组:不运动组(NE)、运动组(EC)和管花肉苁蓉醇提物低、中、高剂量组(E+CtW-L、E+CtW-I和E+CtW-H);小鼠连续给药14 d后,采用10阶段性力竭跑台运动法制备运动性疲劳模型;小鼠运动至力竭状态后处死并采集血清、心、肝、脾、肺、肾以及股骨部肌肉和胃肠样品,测定运动疲劳相关生化指标并制备肺组织切片。结果显示,与NE组相比,EC组小鼠第9阶段出现疲劳状态,其血清尿素氮(UREA)、肌酸激酶(CK)含量均极显著升高(P<0.01),而血糖(GLU)、乳酸脱氢酶(LDH)、肝糖原(HG)、肌糖原(MG)含量均极显著降低(P<0.01),乳酸(LAC)含量也有升高的趋势(P=0.099)。各CtW组小鼠运动至第9阶段未出现明显疲劳状态,各项血清生化指标均维持在与NE组一致的水平。当小鼠继续运动至第10阶段达到力竭状态时,与EC组相比,各CtW组的运动时间和距离均极显著延长(P<0.01);各CtW组的LDH和CK含量、E+CtW-L组和E+CtW-I组的UREA含量以及E+CtW-I组的MG和HG含量与NE组相比无显著差异(P>0.05)。结果表明管花肉苁蓉具有抗小鼠运动性疲劳的作用。

【Abstract】 This study evaluated the effect of Cistanche tubulosa Wight(CtW) on exercise-induced fatigue in mice. Sixty healthy male Kunming mice were randomly divided into 5 groups, namely non-exercise control group(NE), exercise control group(EC), and exercise with supplement of CtW in low-, intermediate-and high-dose groups(E+CtW-L, E+CtW-I and E+CtW-H). After 14 days oral administration of CtW, mice had a 10-stage exercise till exhaustion on treadmill. After exercised to exhaustion, the mice were sacrificed and serum, heart, liver, spleen, lung, kidney, femur muscle and gastrointestinal samples were collected for biochemical testing, and lung tissue sections were prepared. The results showed that compared to those in NE group, mice in EC group had obvious symptom of exercise-induced fatigue in stage 9, with significantly elevated levels of serum urea nitrogen(UREA), creatine kinase(CK)(P<0.01), significantly decreased levels of blood glucose(GLU), lactate dehydrogenase(LDH), hepatic glycogen(HG) and muscle glycogen(MG)(P<0.01), but increased lactose(LAC)(P=0.099); however, mice in CtW groups displayed no obvious symptom of exercise-induced fatigue in stage 9, and all serum biochemical indexes were maintained at the same level as those in NE group. When mice continued to run till exhaustion in stage 10, the exercise duration and distance in CtW groups was significantly prolonged than that in EC group(P<0.01); LDH and CK in all CtW groups, UREA in E+CtW-L and E+CtW-I groups as well as MG and HG in E+CtW-I group were not different from those in NE group(P>0.05). Thus, CtW provides relief on exercise-induced fatigue in mice.
