陕西省及甘肃省部分地区山羊和绵羊干酪性淋巴结炎样脓肿中病原菌种类的调查与分析
Investigation and analysis of pathogenic bacteria in goats and sheep caused by caseous lymphadenitis abscess from Shaanxi and Gansu Provinces
【摘要】 本试验在陕西省关中地区的8家山羊养殖场和甘肃省庆阳地区的14家绵羊养殖场中分别调研11 560只山羊和12 863只绵羊,无菌采集山羊和绵羊体表脓肿样品135份和107份,对羊群临床发病率进行统计,并对采集的样品进行细菌分离培养和16S rRNA基因PCR扩增及测序。结果显示,陕西省关中地区的山羊干酪性淋巴结炎临床发病率为0.3%~9.5%,甘肃省庆阳地区的绵羊干酪性淋巴结炎临床发病率为0.2%~10.1%,平均发病率分别为1.8%和1.9%;细菌分离鉴定结果显示,山羊和绵羊体表脓肿样品中,伪结核棒状杆菌分离率分别为31.3%(42/135)和0.0%(0/107),化脓隐秘杆菌分离率分别为11.9%(16/135)和20.6%(22/107),金黄色葡萄球菌分离率分别为51.1%(69/135)和76.6%(82/107),其他细菌的分离率分别为10.4%(14/135)和5.6%(6/107),而山羊和绵羊样品中分离到单一种类细菌的占比分别为86.7%(117/135)和92.5%(99/107);对同1只羊体表有多处脓肿的样品进行细菌分离鉴定,结果显示山羊体表每处脓肿的病原菌种类既可不一致也可一致,而绵羊的每处脓肿病原菌均表现为一致。
【Abstract】 We investigated 11 560 goats from 8 goat farms in Guanzhong area of Shaanxi Province and 12 863 sheep from 14 sheep farms in Qingyang area of Gansu Province, collected 135 samples and 107 samples from goats and sheep respectively.We calculated the incidence rate by counting the number of diseased ovine and healthy ovine, the collected samples were subjected to bacterial isolation, the isolates were identified by PCR and sequence determination of 16 S rRNA gene.The results showed that the clinical incidence rate of goats caseous lymphadenitis was 0.3%-9.5%,the clinical incidence rate of sheep was 0.2%-10.1%,with an average incidence rate of 1.8% and 1.9% respectively.The results of bacterial isolation and identification showed that the isolation rate of Corynebacterium pseudotuberculosis,Trueperella pyogenes,Staphylococcus aureus and other bacteria in goats and sheep were 31.3%(42/135) and 0(0/107),11.9%(16/135) and 20.6%,51.1%(69/135) and 76.6%(82/107),10.4%(14/135) and 5.6%(6/107),respectively.Among them, the proportion of single bacteria isolated from goats and sheep samples was 86.7%(117/135) and 92.5%(99/107).The results of bacteria identification of individual bovine which appeared multiple abscesses on body surface showed that the pathogenic bacteria in each abscess on the goats body surface could be inconsistent or consistent, while the sheep were all consistent.
