猪细小病毒非结构蛋白NS1通过NF-κB信号通路介导炎症反应
Porcine parvovirus nonstructural protein NS1 inducing inflammatory through NF-κB signal pathway
【摘要】 为了探究猪细小病毒(porcine parvovirus, PPV)非结构蛋白NS1介导炎症反应的情况,将PPV NS1重组质粒转染至PK-15细胞,应用实时荧光定量PCR和免疫印迹技术分别检测炎性因子IL-6/TNF-α的表达和NF-κB信号通路相关蛋白的激活。结果显示,PPV NS1极显著诱导IL-6和TNF-αmRNA的表达(P<0.01)。PPV NS1可显著促进IκBα的降解、p65的磷酸化来激活NF-κB信号通路(0.01<P<0.05)。此外,PPV NS1还促进了Toll样受体2(toll-like receptor 2,TLR2)的表达,TLR2的激动剂和抑制剂分别极显著激活或抑制PPV NS1诱导的IL-6和TNF-α的表达(P<0.01)。结果表明,PPV NS1通过TLR2激活NF-κB信号通路上调炎性细胞因子IL-6和TNF-α的表达。
【Abstract】 To investigate the activation of inflammatory response by the non-structural protein NS1 of porcine parvovirus(PPV),PPV NS1 recombinant plasmid was transfected into PK-15 cells.Real-time quantitative PCR and Western blot were used to detect the expression of inflammatory factor and NF-κB pathway-related protein.Our findings demonstrated that PPV NS1 could induce the mRNA expression of IL-6 and TNF-α(P<0.01).In addition, PPV NS1 promoted the degradation of IκBα and phosphorylation of p65(0.01<P<0.05).PPV NS1 also promoted the expression of toll-like receptor 2(TLR2).At the same time, the agonist and inhibitor of TLR2 showed TLR2 plays an important role in the activation of NF-κB signal pathway inducing by PPV NS1(P<0.01).In conclusion, these results demonstrate that PPV NS1 induces up-regulation of inflammatory cytokines via TLR2-activated NF-κB signaling pathway.These findings provide a new way to study the innate immune mechanism of PPV infection.
