贵州地区人类埃可病毒14型的VP1基因特征分析

【摘要】 目的 分析贵州地区人类埃可病毒14型(Echovirus 14,ECHO 14)分离株的VP1基因特征,为ECHO 14的防控提供科学依据。方法 收集2001—2021年贵州省急性弛缓性麻痹(acute flaccid paralysis, AFP)监测系统中从AFP病例及健康儿童粪便标本中分离到的15株ECHO 14毒株并进行回顾性鉴定分析,采用逆转录-聚合酶链反应(RT-PCR)扩增出VP1基因并对扩增产物测序,用MEGA7.0构建系统进化树并对氨基酸位点突变进行分析。结果 贵州地区15株ECHO 14分离株属于A、B和C基因亚型,其中A基因亚型占比达80.0%(12/15)。15株ECHO 14分离株之间的核苷酸一致性为81.8%~97.1%,氨基酸同源性为94.3%~99.7%,与原型株Tow (Rhode Island/54)株之间的核苷酸一致性为77.1%~79.4%,氨基酸同源性为95.1%~98.4%;与ECHO 14原型株Tow株比较分析,结果显示在VP1氨基酸序列上存在33个突变位点,其中8个位点已完全突变分别为N8D、K132T、G157D、L217M、V237I、M295L、H296Q和H298R。结论 2000—2021年贵州省AFP病例中分离的ECHO 14毒株属于A、B和C基因亚型,VP1区的氨基酸多位点变异,可能导致了多条传播链在贵州地区共循环,其中A基因亚型是2006年后贵州地区传播的优势亚型。

【Abstract】 Objective To understand the VP1 genetic characteristics of Echovirus 14(ECHO 14) strains isolated in Guizhou province of China,and to provide scientific basis for prevention and control of ECHO14.Methods Fifteen strains of ECHO14 isolated from stool samples of cases with acute flaccid paralysis(AFP)and healthy children in the surveillance system of AFP in Guizhou during 2001 and 2021 were collected and retrospectively analyzed.The VP1 gene was amplified by reverse transcription-polymerase chain reaction(RTPCR) and the amplified products were sequenced.The evolutionary tree was constructed by MEGA7.0 and the amino acid site mutation was analyzed.Results Fifteen ECHO 14 isolates in Guizhou belonged to A,B and C subtypes.Among them,genotype A accounts for 80.0%(12/15).The nucleotide and the amino acid homologies were 81.8%—97.1 % and 94.3%—9.7% among 15 ECHO 14 isolates,and were 77.1%—79.4% and95.1 %—98.4% compared with the ECHO 14 prototype(Tow,Rhode Island/54).Compared with ECHO 14prototype strain Tow,there were 33 mutation sites in VPl amino acid sequence,of which 8 sites were completely mutated as N8D,K132T,G157D,L217M,V237I,M295L,H296Q and H298R,respectively.Conclusions ECHO 14 strains isolated from AFP cases in Guizhou province from 2000 to 2021 belong to A,B and C genotypes.The amino acid multi-site variation in VP1 region may lead to the co-circulation of multiple transmission chains in Guizhou,among which A subtype becomes the dominant population in Guizhou after 2006.
