肠致病性大肠杆菌紧密黏附素的作用机制及基因分型研究进展
Research progress on the adhesion mechanism and genotyping of intimin in enteropathogenic Escherichia coli
【摘要】 肠致病性大肠杆菌(Enteropathogenic Escherichia coli, EPEC)引起的腹泻性疾病是世界范围内重要的公共卫生问题。在EPEC黏附肠道细胞的过程中,紧密黏附素与易位受体Tir相互作用触发了肌动蛋白的组装,在细菌与宿主细胞连接部位形成了紧密附着的基座。紧密黏附素不仅是EPEC产生特征性黏附/消除病变的关键因子,还对宿主具有不同的组织向性。编码紧密黏附素的eae基因是鉴定EPEC菌株的靶点,其序列存在相当大的异质性,是EPEC分型的重要遗传学基础。同时,携带不同eae亚型的LEE致病岛可能在不同血清型、不同类型的致泻性大肠杆菌菌株之间发生水平转移。因此,在预测EPEC腹泻性流行病的严重程度时,可对EPEC菌株的eae基因变异做鉴定和分型,为监测方向提供参考。
【Abstract】 Diarrheal diseases caused by enteropathogenic Escherichia coli(EPEC) pose a challenge to public health worldwide. During the adhesion of EPEC to intestinal cells, the interaction of intimin with the translocation receptor Tir triggers the assembly of actin, forming a tightly adherent base at the site of bacterial-host cell attachment. Intimin is not only an essential key factor allowing EPEC to produce characteristic attaching-and-effacing lesions, but also has different tissue orientation to the host. The intimin encoding gene eae, a target for identifying EPEC strains, has considerable heterogeneity and is an important genetic basis for EPEC typing. Moreover, LEE pathogenicity islands carrying different eae subtypes may be transferred horizontally among different serotypes of diarrheagenic E. coli strains. Thus, identification and typing of the eae gene of EPEC strains could provide a reference for surveillance and prediction of severity in EPEC diarrheal epidemics.
