基于UHPLC-LTQ-Orbitrap MS~n的苦碟子醇提物不同洗脱部位化学成分分析

【摘要】 目的 利用超高效液相色谱-线性离子阱-串联静电场轨道阱质谱(UHPLC-LTQ-Orbitrap MSn)对苦碟子药材醇提物不同洗脱部位的化学成分进行分析鉴定。方法 采用Acquity UHPLC BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),以乙腈(A)-0.1%甲酸-水溶液(B)为流动相进行梯度洗脱,在负离子模式下,分别对苦碟子药材醇提物经大孔吸附树脂分离的30%、50%、70%和95%乙醇洗脱部位进行数据采集。根据高分辨质谱提供的准分子离子峰和多级质谱碎片,获取目标化合物的精确分子量和结构信息,并结合保留时间、ClogP值、对照品分析、参考文献等相关数据对化合物进行鉴别。结果 分别在30%、50%、70%和95%乙醇洗脱部位中鉴定出36、56、33和15个化合物,包括核苷类、黄酮类、有机酸类、倍半萜内酯类、三萜类、苯丙素类等共80个化学成分,其中5个为共有成分,分别为木犀草素、芹菜素、木犀草素-7-O-β-D-葡萄糖苷、芹菜素-7-O-β-D-葡萄糖苷和七叶内酯。结论 该研究较为全面地表征了苦碟子药材醇提物的化学成分基础,并分析了其不同洗脱部位化学成分的差异,为进一步阐明苦碟子的药效物质基础提供了科学依据。

【Abstract】 Objective To determine the chemical components of Ixeris sonchifolia(Bunge)Hance alcohol extract by ultra-high performance liquid chromatography equipped with linear ion trap-orbitrap mass spectrometry (UHPLC-LTQ-Orbitrap MSn).Methods An Acquity UHPLC BEH C18 column (2.1 mm×100 mm,1.7 μm) was used for the gradient elution with acetonitrile and water (0.1% formic acid).Different elution parts separated by macroporous resin column from Ixeris sonchifolia(Bunge)Hance alcohol extract were analyzed in a negative ion mode.Components of Ixeris sonchifolia(Bunge)Hance alcohol extract were identified by the mass spectra.The chromatographic behaviors of the compounds were compared,as well as the retention time,ClogP,accurate mass measurements and mass fragmentation patterns.Results Totally 80 compounds were identified from Ixeris sonchifolia(Bunge)Hance alcohol extract.These compounds covered nucleoside,flavonoids,organic acids,sesquiterpene lactones,triterpenoids and phenylpropanoids.Totally 36,56,33 and 15 constituents were identified from 30%,50%,70% and 95% ethanol,respectively.Five common components (luteolin,apigenin,luteolin-7-O-β-D-glucoside,apigenin-7-O-β-D-glucoside and aesculolide) were shown in different elution parts.Conclusion The chemical constituents of Ixeris sonchifolia(Bunge)Hance alcohol extract are characterized,and the difference in the chemical constituents of different elution parts is analyzed,which provides scientific data for elucidating medicinal materials of Ixeris sonchifolia(Bunge)Hance.
