不同等级高温大曲微生物群落和代谢物的特征分析

【摘要】 该研究采用高通量测序(HTS)、气相色谱-离子迁移谱(GC-IMS)和氢谱核磁共振(1H NMR)比较了不同等级高温大曲微生物群落和代谢物的差异。HTS结果表明,不同等级高温大曲基于属水平的优势菌群有一定差异,一级大曲中优势菌属为慢生芽孢杆菌属(Lentibacillus)、克罗彭斯特菌属(Kroppenstedtia)、葡萄球菌属(Staphylococcus)和曲霉属(Aspergillus)等,优级大曲中优势菌属为芽孢杆菌属(Bacillus)、克罗彭斯特菌属和踝节菌属(Rasamsonia)等,特级大曲中优势菌属为芽孢杆菌属、克罗彭斯特菌属和曲霉属等。GC-IMS结果表明,不同等级高温大曲挥发性代谢物组成及含量的差异主要体现在醇类、醛酮类和酯类等物质。1H NMR检测结果表明,不同等级高温大曲中非挥发性代谢物组成及含量的差异主要体现在氨基酸等物质,且特级大曲非挥发性代谢物的种类更为丰富、含量更高。

【Abstract】 The differences of microbiotas and metabolites in high-temperature Daqu with different grades were compared using high-throughput sequencing(HTS), gas chromatography-ion mobility spectrometry(GC-IMS) and1H nuclear magnetic resonance(1H NMR). The HTS results showed that the dominant community of high-temperature Daqu with different grades had obvious differences based on genus level. The dominant genera were Lentibacillus, Kroppenstedtia, Staphylococcus and Aspergillus in the first-grade Daqu, Bacillus, Kroppenstedtia and Rasamsonia in premium-grade Daqu, and Bacillus, Kroppenstedtia and Aspergillus in super premium-grade Daqu. The GC-IMS results showed that the composition and contents of volatile metabolites were different in high-temperature Daqu with different grades, which were reflected in the content of alcohols, aldehydes, ketones and esters, etc. The1H NMR detection results showed that the differences in the composition and contents of non-volatile metabolites in high-temperature Daqu with different grades were mainly reflected in the content of amino acids, etc., and the species and contents of non-volatile metabolites of super premium-grade Daqu were more abundant and higher.
