直肠癌患者PANDAR mRNA表达与临床病理特征及预后的关系

【摘要】 目的 探讨直肠癌患者PANDAR mRNA表达与临床病理特征及预后的关系。方法 选取2015年1月—2016年12月在中国科学院大学附属肿瘤医院接受直肠癌根治术的120例患者为研究对象,比较直肠癌组织与癌旁正常组织中PANDAR mRNA表达水平,以及直肠癌组织PANDAR mRNA高、低表达患者临床病理特征及预后,采用多因素logistic回归分析直肠癌患者PANDAR m RNA表达的影响因素。结果 直肠癌组织中PANDAR mRNA表达水平明显高于癌旁正常组织(P<0.05)。PANDAR mRNA高表达(表达水平>5.25)65例,低表达(表达水平≤5.25)55例。直肠癌组织PANDAR mRNA高、低表达患者在远处转移、淋巴结转移、肿瘤浸润深度、MRI-T分期、MRI-N分期等方面比较,差异均有统计学意义(均P<0.05);在性别、年龄、肿瘤直径、肿瘤组织分化、MRI-环周切缘状态等方面比较,差异均无统计学意义(均P>0.05)。MRI-T分期(OR=6.071,95%CI:1.281~28.783,P<0.05)、MRI-N分期(OR=12.481,95%CI:2.046~76.148,P<0.05)是直肠癌患者PANDAR mRNA表达的独立影响因素。术后随访48个月,PANDAR mRNA低表达患者4年总体生存率明显高于PANDAR mRNA高表达者,差异有统计学意义(39.2%比19.8%,P<0.05)。结论 PANDAR在直肠癌组织中呈高表达,与MRI-T分期、MRI-N分期等临床病理特征及预后相关。

【Abstract】 Objective To investigate the expression of lncRNA PANDAR in rectal cancer and its relation with clinicopathological features and prognosis of patients. Methods One hundred and twenty patients with rectal cancer who underwent radical resection in Cancer Hospital to University of Chinese Academy of Sciences from January 2015 to December 2016 were enrolled in the study. The expression of lncRNA PANDAR were detected in cancer tissues and pericancerous tissues, and the relationship of lncRNA PANDAR expression with clinicopathological characteristics and prognosis of patients was analyzed. Results The expression level of PANDAR mRNA in rectal cancer tissues was significantly higher than that in pericancerous tissue(P<0.05). There were 65 cases with high PANDAR expression level(>5.25)and 55 cases with low expression level(≤5.25). The distant metastasis, lymph node metastasis, depth of tumor infiltration,MRI-T stage, MRI-N stage in patients with high PANDAR expression level were higher than those with low expression level(all P<0.05); while there were no significant differences in gender, age, tumor diameter, tumor tissue differentiation, MRIperipheral cutting edge status between high and low expression groups(all P>0.05). MRI-T stage(OR=6.071, 95%CI: 1.281-28.783, P<0.05) and MRI-N stage(OR=12.481, 95% CI: 2.046-76.148, P<0.05) were independent factors in predicting PANDAR expression in rectal cancer patients(P<0.05). Patients were followed up for 48 months after surgery, and the overall survival rate of patients with low PANDAR expression was significantly higher than that with high PANDAR expression(39.2% vs. 19.8%, P<0.05). Conclusion lncRNA PANDAR is highly expressed in rectal cancer, which is correlated with depth of invasion, distant metastasis, lymph node metastasis, MRI-T stage, MRI-N stage, and the prognosis of patients.
