藜麦EMS诱导条件优化及穗突变体的筛选

【摘要】 通过不同浓度EMS(0.5%、1.0%、1.5%、2.0%、2.5%、3.0%)对4个藜麦品系进行不同时长(4 h、6 h、8 h、10 h、12 h)诱变处理,构建了5 000株白藜麦突变群体。研究表明,随着EMS浓度从0.5%增至3.0%,发芽率从90%降至50%,成苗率降至10%以下。随着诱变剂EMS处理浓度的增加和处理时间的延长,藜麦种子的发芽势和成苗率均呈现下降趋势,抑制作用明显;EMS对种子萌发的影响小于幼苗。获得EMS处理藜麦种子的最适处理浓度为1.5%,处理时间为6 h,种子萌发率为80%,成苗率下降到50%。本试验最终筛选出4株穗型变异的突变体植株。

【Abstract】 In this study, four varieties of Chenopodium quinoa Willd. were treated with combination of different ethyl methanesulfonate(EMS) concentrations(0.5%, 1.0%, 1.5%, 2.0%, 2.5%, 3.0%) and treatment time of 4 h, 6 h, 8 h, 10 h and 12 h, and a mutant group of 5 000 strains of white Chenopodium quinoa Willd. was constructed. The study showed that the germination rate decreased from 90% to less than 50% and the seedling rate decreased to less than 10% while the EMS concentration increased from 0.5% to 3.0%. With the increase of EMS treatment concentration and treatment time, the germination potential and seedling rate of Chenopodium quinoa Willd. seeds showed a downward trend, showing an obvious inhibitory effect. The effect of EMS on seed germination was less than that of seedlings. The suitable treatment of Chenopodium quinoa Willd. seeds treated by EMS was 1.5% for 6 h, resulting in 80% germination rate and 50% seedling rate. Finally, four mutant plants with spike type variation were screened out.
