油莎草提取物对慢性应激诱导小鼠抑郁样行为的改善和作用机制初探

【摘要】 目的:研究油莎草提取物对慢性温和不可预见性应激(CUMS)小鼠的抗抑郁功效及机制。方法:采用CUMS程序持续6 w建立抑郁模型,造模成功后,随机将小鼠分为模型组、阳性组、油莎草须根水提物剂量组、油莎草须根醇提物剂量组、油莎草地上茎叶水提物剂量组、油莎草地上茎叶醇提物剂量组、油莎豆水提物剂量组、油莎豆醇提物剂量组,每组14只。灌胃给药,持续30 d,模型组和正常组灌胃等体积的双蒸水,阳性组氟西汀按20 mg/(kg·d)BW灌胃,其余6组皆按200 mg/(kg·d)BW灌胃。除正常对照组外,其余组均采用CUMS小鼠模型,并观察油莎草提取物对小鼠行为学、生化指标和皮质酮等指标的影响。每周进行体重称量,采用旷场实验、强迫游泳实验、糖水偏好实验等行为学方法评价小鼠的抑郁状态;采用试剂盒法测定小鼠血清MDA含量、SOD、GSH-PX活性等生化指标,采用酶联免疫分析(ELISA)法测定小鼠血清皮质酮含量。结果:与正常组比较,模型组小鼠生长缓慢,运动功能明显受到抑制,小鼠水平、垂直运动得分降低(P<0.01),游泳不动时间明显延长,糖水相对消耗量明显降低(P<0.01);血清SOD、GSH-Px活性降低,MDA含量明显增高(P<0.01),皮质酮水平明显升高(P<0.01);与模型组比较,油莎草提取物各组小鼠生长正常,小鼠水平、垂直运动得分提高(P<0.01),游泳不动时间显著缩短(P<0.01),蔗糖水相对消耗量明显升高(P<0.01);血清SOD、GSH-Px活力提高,MDA含量明显降低(P<0.01),小鼠血清皮质酮水平明显降低(P<0.01);与正常组比较无统计学差异。结论:油莎草提取物对慢性应激所引起的抑郁症状有一定的改善功效,其作用机制可能与抑制HPA轴功能紊乱,改善机体自由基损伤状态,增强SOD、GSH-Px活力,降低MDA含量等多种途径有关。

【Abstract】 Objective To investigate the effect and mechanism of Cyperus esculentus( CEL) extracts on chronic unpredictable mild stress( CUMS) induced depression mice. Method The depression model were established by the procedure of chronic unpredictable mild stress stimulation( CUMS) combined with lonely raising for 42 days. Depressed mice were randomly divided into model group,positive group,CEL fibrous root water extract dose group,CEL fibrous root alcohol extract dose group,CEL stem and leaf on the ground water extract dose group,CEL water extract dose group and CEL alcohol extract dose group,with 14 mice in each group. The drugs were given by gavage once a day for consecutive 30 days. Model group and normal group were given equal volume of double steam water,positive fluoxetine group was given 20 mg/( kg·d) BW,the other 6 groups were given 200 mg/( kg·d) BW. Except the normal control group,the CUMS mice were used in other groups,and the effects of CEL extract on behavior,biochemical indices and corticosterone were observed. The mice were weighed weekly,and the depression status of mice was evaluated by observing open field test,forced swimming test and sugar water preference test. The activity of superoxide dismutase( SOD) and glutathione peroxidase( GSH-PX) and the content of malondialdehyde(MDA) were measured by kit method. The content of corticosterone in serummicewere determined by ELISA. Result Compared with control group,CUMS mice grew slowly. The motor function was significantly injured,the scores of horizontal and vertical movement were lowered. The time of motionless swimming was prolonged significantly,and the consumption of sugar water was significantly decreased. The activities of SOD and GSH-Px in serum decreased while the content of MDA significantly increased,and the corticosterone levels were significantly elevated,Compared with model group,each of the CEL extract groups grew normally,the horizontal and vertical movement scores of mice were improved,the immobility time of forced swimming test was significantly shortened and sugar water consumption were significantly increased. The activities of SOD and GSH-Px in serum increased while the content of MDA significantly decreased,and the serum corticosterone level of mice decreased significantly. In addition,there was no statistical difference compared with normal group. Conclusion CEL extract has a prominent therapeutic effect on mice model of chronic stress-induced depression and the mechanism might be related with reduction of the hyperfunction of hypothalamus-pituitary-adrenal( HPA),improvement of free radical damage,enhancement of the activity of SOD and GSH-Px,reduction of the content of MDA.
