低强度脉冲超声靶向辐照大鼠脾脏对免疫应答的影响

【摘要】 目的 探讨低强度脉冲超声(LIPUS)靶向辐照斯泼累格·多雷(SD)大鼠脾脏对免疫应答的影响及其相关机制。方法 42只SD大鼠随机分成2组:超声组(LIPUS,n=21)和对照组(Control,n=21)。LIPUS靶向辐照脾脏7、14和21 d后,采用酶联免疫吸附测定试验(ELISA)检测免疫球蛋白A(IgA)、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、白细胞介素2(IL-2)、白细胞介素7(IL-7)、白细胞介素10(IL-10)、肿瘤坏死因子-α(TNF-α)和γ干扰素(IFN-γ)水平;采用免疫组化(IHC)观察TNF-α和IFN-γ在脾脏及胸腺中的表达水平。结果 与Control组相比,LIPUS组血浆中IgM及免疫因子IL-2、IL-7、IL-10、TNF-α水平显著升高(P<0.05),两组IgA和IgG的表达水平在各时间点的差异无统计学意义;免疫器官脾脏中IL-7、IL-10、TNF-α和IFN-γ水平上调以及胸腺中IL-7、IL-10的表达增高,明确了LIPUS对调节大鼠免疫因子及改善其免疫力的作用。结论 LIPUS靶向作用于脾脏可以改善机体免疫球蛋白水平,诱导免疫因子的释放,从而增强机体的免疫力。

【Abstract】 Objective To investigate the effects of immune response in Sprague-Dawley(SD) rat with low-intensity pulsed ultrasound(LIPUS) targeted irradiation on spleen and its related mechanisms. Methods Forty-two SD rats were randomly divided into 2 groups: Ultrasound group(LIPUS, n=21) and control group(Control, n=21). Immunoglobulin A(IgA), immunoglobulin G(IgG), immunoglobulin M(IgM), interleukin 2(IL-2), interleukin 7(IL-7), interleukin 10(IL-10), tumor necrosis factor-α(TNF-α) and gamma interferon(IFN-γ) levels were measured by enzyme-linked immunosorbent assay after 7, 14 and 21 d of LIPUS-targeted spleen irradiation, and the expression of TNF-α and IFN-γ in the immune organs of spleen and thymus was observed by immunohistochemistry. Results Plasma levels of IgM and immune factors IL-2, IL-7, IL-10, TNF-α were significantly higher in the LIPUS group compared with the Control group(all P<0.05), while the differences in the expression levels of IgA and IgG between the two groups at each time point were not statistically significant. The levels of IL-7, IL-10, TNF-α and IFN-γ were upregulated in the immune organ spleen and the expression of IL-7, IL-10 was increased in the thymus, which clarified the effects of LIPUS on regulating immune factors and improving immunity in rats. Conclusions Targeted action of LIPUS on the spleen can improve the immunoglobulin levels and induce the release of immune factors, thus enhancing the immunity of the body.
