自噬相关基因在胃癌组织中的表达及其临床意义研究
ATG expression and clinical significance in gastric cancer
【摘要】 目的 探讨自噬相关基因(autophagy associated gene, ATG)ATG 7、 ATG 10、 ATG 12、 Beclin-1在胃癌组织中的表达情况及临床意义。方法选取2018年8月~2020年6月本院收集的45例胃癌组织和对应的癌旁组织作为研究对象。采用荧光定量聚合酶联反应(PCR)分析癌旁组织和胃癌组织中自噬相关基因m RNA的表达,同时采用免疫组织化学染色(IHC)分析癌旁组织和胃癌组织中自噬相关基因蛋白表达情况。计数资料采用率(%)表示,组间比较采用t检验,组间相关性采用one-way ANOVAs分析。结果 45例胃癌组织、相对应的癌旁组织及10例正常对照中的ATG表达情况显示,胃癌组织ATG的表达与癌旁组织相比较,二者差异无统计学意义(P> 0.05);胃癌组织、癌旁组织与正常对照组比较,二者差异均有统计学意义(P <0.05)。结论 ATG 7、 ATG 10、 ATG 12、 Beclin-1基因在胃癌发生发展过程中发挥一定作用,联合检测前述自噬相关基因在制定抗肿瘤治疗策略中有望成为有效的靶点。
【Abstract】 Objective To investigate the expression of the ATG(autophagy associated gene),including ATG 7、 ATG 10、 ATG 12、 Beclin-1 in gastric cancer, as well as its clinical significance. Methods 45 gastric cancer pathological specimens from surgery in our hospital from August 2018 to June 2020 were collected, together with appropriate 45 adjacent normal gastric mucosa tissues. Related mRNA expression was determined by RT-qPCR and autophagy associated protein was detected by immunohistochemistry in 45 pairs of gastric tumors corresponding non-neoplastic gastric tissue and 10 samples of matched non-neoplastic gastric tissue. Paired t-tests were performed to compare the mean these genes expression between tissue adjacent to tumors non-neoplastic and tumor samples.The associations between clinicopathological parameters and the mean of ATG genes expression was assessed using t-tests for independent samples and one-way ANOVAs. Results The expression of ATG in 45 gastric cancer tissues, corresponding adjacent tissues and 10 normal controls showed that there was no significant difference between the expression of ATG in gastric cancer tissues and adjacent tissues(P > 0.05); There were significant differences between gastric cancer tissue and normal control group,adjacent tissue and normal control group(P < 0.05). Conclusion The results showed that ATG7、ATG10、ATG 12 and Beclin-1gene may act as a tumor suppressor in gastric carcinogenesis and may help in the selection of anticancer treatment strategies.
