额日赫木-8散HPLC指纹图谱建立及秦皮乙素测定

【摘要】 目的 建立额日赫木-8散(红花、黄连、紫花地丁等)HPLC指纹图谱,并测定秦皮乙素含量。方法 分析采用Agilent SB C18或πNAP COSMASIL色谱柱(4.6 mm×250 mm, 5μm);流动相甲醇-0.1%磷酸,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长345 nm。聚类分析、主成分分析、偏最小二乘法判别分析评价批间质量差异。结果 10批样品指纹图谱中有17个共有峰,相似度均大于0.9。各批样品聚为3类,4、10、12、15号峰是主要差异标志物。秦皮乙素在0.414 1~106μg/mL范围内线性关系良好(r=0.999 9),平均加样回收率为96.3%,RSD为0.42%。结论 该方法简便准确,重复性好,可用于额日赫木-8散的质量控制。

【Abstract】 AIM To establish the HPLC fingerprints for Erihemu-8 Powder(Carthami Flos, Coptidis Rhizoma, Violae Herba, etc.) and to determine the content of aesculetin.METHODS The analysis was performed on a 30 ℃ Agilent SB C18 or πNAP COSMASIL C18 column(4.6 mm×250 mm, 5 μm), with the mobile phase comprising of methanol-0.1% phosphoric acid flowing at 1.0 mL/min in a gradient elution manner, and the detection wavelength was set at 345 nm. Cluster analysis, principal component analysis and partial least squares discriminant analysis were applied to evaluating quality difference between batches. RESULTS There were seventeen common peaks in the fingerprints of ten batches of samples with the similarities of more than 0.9. Various batches of samples were clustered into three types, peaks 4, 10, 12 and 15 were main difference markers. Aesculetin showed a good linear relationship within the range of 0.414 1-106 μg/mL(r=0.999 9), whose average recovery was 96.3% with the RSD of 0.42%.CONCLUSION This simple, accurate and reproducible method can be used for the quality control of Erihemu-8 Powder.
