芦根化学成分及其抗氧化和α-葡萄糖苷酶抑制活性
Chemical constituents from Phragmites communis and their antioxidant and α-glucosidase inhibitory activities
【摘要】 目的 研究芦根Phragmites communis Trin.的化学成分及其抗氧化和α-葡萄糖苷酶抑制活性。方法 芦根乙醇提取物采用硅胶、聚酰胺、Sephadex LH-20、反相ODS柱色谱,制备液相色谱等进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用DPPH法评价其抗氧化活性,PNPG法评价其α-葡萄糖苷酶抑制活性。结果 从中分离得到25个化合物,分别鉴定为(+)-南烛木树脂酚9′-O-β-D-葡萄吡喃糖苷(1)、(-)-南烛木树脂酚9′-O-β-D-葡萄吡喃糖苷(2)、(-)-南烛木树脂酚9-O-β-D-葡萄吡喃糖苷(3)、(-)-7′-表-南烛木树脂酚9′-O-β-D-葡萄吡喃糖苷(4)、(-)-8′-表南烛木树脂-4-烯基β-葡萄吡喃糖苷(5)、(+)-南烛木树脂-4-烯基β-D-葡萄吡喃糖苷(6)、丁香脂素4′-O-β-D-葡萄吡喃糖苷(7)、异甘草苷(8)、6″-O-乙酰化甘草苷(9)、甘草苷(10)、甘草查尔酮A(11)、小麦黄素(12)、苜蓿素-7-O-芸香糖苷(13)、苜蓿素-7-O-新橙皮糖苷(14)、2,3-二羟基-1-(4-羟基-3,5-二甲氧基苯基)-1-丙酮(15)、(7R,8S)-愈创木基甘油(16)、(7S,8S)-愈创木基甘油(17)、对羟基桂皮酸(18)、反式阿魏酸(19)、(E)-3,3′-二甲氧基-4,4′-二羟基二苯乙烯(20)、丁香醛(21)、西米杜鹃醇(22)、6′-O-乙酰化-β-胡萝卜苷(23)、果糖(24)、正丁基-O-β-D-吡喃果糖苷(25)。化合物1~4和7抗氧化活性IC50值分别为(60.01±1.12)、(58.40±0.31)、(65.27±1.33)、(65.27±1.33)、(63.77±0.61)μmol/L;化合物2~4、11和15α-葡萄糖苷酶抑制活性IC50值分别为(850.22±2.31)、(847.99±1.84)、(847.99±1.84)、(180.42±0.91)、(1 536.56±1.89)μmol/L。结论 化合物2~11、13~17、20、23~25均为首次从该植物和该属植物中分离得到。化合物1~4和7具有较强的抗氧化活性,化合物2~4、11和15具有α-葡萄糖苷酶抑制活性。
【Abstract】 AIM To study the chemical constituents of Phragmites communis Trin. and their antioxidant and α-glucosidase inhibitory activities. METHODS The ethanol extract from P. communis was isolated and purified by silica, Sephadex LH-20, ODS,and preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data. DPPH method and PNPG method was used to evaluate their antioxidant and α-glucosidase inhibitory activities. RESULTS Twenty-five compounds were isolated and identified as(+)-lyoniresinol 9′-O-β-D-glucopyranoside(1),(-)-lyoniresinol 9′-O-β-D-glucopyranoside(2),(-)-lyoniresinol 9-O-β-D-glucopy-ranoside(3),(-)-7′-epi-lyoniresinol 9′-O-β-D-glucopyranoside(4),(-)-8′-epilyoniresin-4-yl β-glucopyranoside(5),(+)-lyoniresin-4-yl β-D-glucopyranoside(6), syringaresinol-4′-O-β-D-glucopyranoside(7), isoliquiritin(8), 6″-O-acetylliquiritin(9), liquiritin(10), licochalcone A(11), tricin(12), tricin-7-O-rutinoside(13), tricin-7-O-neohesperidoside(14), 2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone(15),(7R,8S)-guaiacyl glycerol(16),(7S,8S)-guaiacyl glycerol(17), p-coumaric acid(18), trans-ferulic acid(19),(E)-3,3′-dimethoxy-4,4′-dihydroxystilbene(20), syringaldehyde(21), simiarenol(22), 6′-O-acetyl-β-daucosterol(23), fructose(24) and n-butyl-β-D-fructopyranoside(25). The antioxidant activity IC50 values of compounds 1-4 and 7 were(60.01±1.12),(58.40±0.31),(65.27±1.33),(65.27±1.33),(63.77±0.61) μmol/L, respectively; and the α-glucosidase inhibitory activity IC50 values of compounds 2-4, 11 and 15 were(850.22±2.31),(847.99±1.84),(847.99±1.84),(180.42±0.91),(1 536.56±1.89) μmol/L, respectively.CONCLUSION Compounds 2-11, 13-17, 20 and 23-25 are isolated from this plant and genus for the first time. Compounds 1-4 and 7 have potent antioxidant activities, and compounds 2-4, 11 and 15 have α-glucosidase inhibitory activities.
