miR-451在雌激素诱导大鼠乳腺增生中的作用机制研究
Study on the mechanism of miR-451 in estrogen-induced cyclomastopathy in rats
【摘要】 目的 探讨微小RNA(miR)-451对乳腺增生上皮细胞雌激素受体(ER)、细胞周期蛋白D1(Cyclin D1)、B淋巴细胞瘤-2基因(Bcl-2)的影响,为进一步阐明乳腺增生发病机制和发现新治疗靶点奠定基础。方法 2020年6月采用随机数字表法将20只大鼠分为乳腺增生组(采用雌、孕激素序贯法建立大鼠乳腺增生模型)和对照组(注射生理盐水),每组10只。采用定量转录聚合酶链反应检测2组大鼠miR-451的表达,Western-blotting检测2组大鼠ER、Cyclin D1、Bcl-2蛋白的表达,并分析miR-451与ER、Cyclin D1、Bcl-2蛋白表达的相关性。结果 对照组大鼠乳腺组织miR-451表达量是乳腺增生组的1.75倍。乳腺增生组大鼠乳腺组织ER、Bcl-2蛋白相对表达明显高于对照组,差异均有统计学意义(P<0.05)。miR-451与ER、Bcl-2蛋白表达呈明显负相关(r=-0.87、-0.79,P<0.05)。结论 miR-451在乳腺增生组织中表达下调,并可能通过增加ER、Bcl-2蛋白的表达影响疾病的发生、发展。
【Abstract】 Objective To investigate the effects of microRNA(miR)-451on the expression levels of estrogen receptor(ER),cell cycle protein D1(Cyclin D1)and B lymphocytoma-2gene(Bcl-2)in proliferative mammary epithelial cells,elucidate the pathogenesis of cyclomastopathy and lay the foundation find for discovering new therapeutic target.Methods In June 2020,a total of 20rats were randomly divided into control group and cyclomastopathy group by the random number table method,with 10rats per group.Rats in cyclomastopathy group were conducted with estrogen and progestogen by using sequential method,rats control group were injected with normal saline.The expression levels of miR-451were detected by using quantitative reverse transcription PCR,western blotting test was used to detect the expression levels of ER,Cyclin D1and Bcl-2protein in two groups of rats.The correlations of miR-451with the expression of ER,Cyclin D1,and Bcl-2protein were also analyzed.Results The expression of miR-451in control group was 1.75times higher than that in cyclomastopathy group,and the relative expression levels of ER,Bcl-2protein were significantly higher than those of control group.There was a negative correlation between the expression levels of miR-451and the relative expression levels of ER and Bcl-2protein(r=-0.87,-0.79,P<0.05).Conclusion The expression levels of miR-451in cyclomastopathy group is down-regulated,and may influence the occurrence and development of cyclomastopathy through increasing the relative expression levels of ER,Bcl-2protein.
