不同条件储存制备新鲜冰冻血浆对质量的影响

【摘要】 目的 分析不同条件储存制备新鲜冰冻血浆的凝血因子Ⅷ活性的变化,探讨血液储存条件、制备时间等因素对新鲜冰冻血浆制备质量的影响。方法 随机选取捐献全血的无偿献血者40名,将采集的全血分为A、B、C、D四组,在储存的8 h、12 h和24 h分别制备新鲜冰冻血浆,并进行凝血因子Ⅷ活性的检测。结果 不同条件储存制备的新鲜冰冻血浆,其凝血因子Ⅷ活性随着制备时间的延长而下降(P<0.05)。各组各种时间段制备新鲜冰冻血浆的凝血因子Ⅷ活性检测结果为:A组>B组>C组>D组;全血在储存24 h后,各组制备新鲜冰冻血浆的凝血因子Ⅷ含量合格率分别为A组100%,B组75%,C组80%,D组65%。结论 新鲜冰冻血浆的凝血因子Ⅷ活性随着制备时间的延长而下降,全血储存24 h内制备FFP的合格率室温条件(20-24℃)优于低温条件(2-6℃)。

【Abstract】 Objective The changes of coagulation factor VIII activity of fresh frozen plasma prepared under different storage conditions were analyzed,and the effects of blood storage conditions,preparation time and other factors on the preparation quality of fresh frozen plasma were discussed.Methods Forty unpaid blood donors who donated whole blood were randomly selected and the collected whole blood was divided into A,B,C and D groups. Fresh frozen plasma was prepared at 8 h,12 h and 24 h of storage,and tested for coagulation factor VIII activity.Results The coagulation factor VIII activity of fresh frozen plasma prepared under different conditions was decreased with the prolongation of the preparation time(P<0.05).The results of coagulation factor VIII activity test of fresh frozen plasma prepared in different groups at different time periods were as follows:group A > group B > group C > group D;After 24 h storage of whole blood,the qualified rates of coagulation factor VIII content of fresh frozen plasma in each group were 100% in group A,75% in group B,80% in group C and 65% in group D,respectively.Conclusion The factor VIII activity of fresh frozen plasma decreased with the preparation time,and the qualified rate of FFP prepared within 24 h after whole blood storage under room temperature(20-24℃)was better than that under low temperature(2-6℃).
