双黄连注射剂中异绿原酸C致敏兔抗体效价测定
Determination of Antibody Titer of Rabbits Induced by Isochlorogenic Acid C in Shuanghuanglian Injection
【摘要】 目的 通过ELISA法测定家兔血清中特异性抗体效价,以此确定双黄连注射剂中异绿原酸C是否具有致敏性。方法 将双黄连注射剂中疑似小分子半抗原物质异绿原酸C与正常家兔血清体外孵育制备完全抗原,免疫家兔获得特异性抗体。采用ELISA法检测抗血清的抗体效价。采用间接竞争ELISA法测定血清特异性并绘制抑制曲线。采用兔免疫球蛋白E(IgE)ELISA试剂盒检测各组家兔抗血清中IgE抗体的含量。结果 异绿原酸C的抗体效价(OD)值高于阴性对照组2倍,说明其具有潜在的致敏性。回归方程I=0.430 lgC+0.319,R2=0.997,半数抑制浓度IC50=2.628 mg·mL-1。家兔体内明显产生了外源性IgE型抗体。结论 在实验室温度为20℃,湿度为40%环境条件下,家兔被给药后制得抗血清,通过ELISA法测定得出双黄连注射剂中疑似半抗原物质异绿原酸C具有致敏性。
【Abstract】 Objective The specific antibody titer in rabbit serum was determined by ELISA to determine whether isochlorogenic acid C in Shuanghuanglian injection has sensitization. Methods The complete antigen was prepared by incubating the suspected small molecular hapten isochlorogenic acid C contained in Shuanghuanglian injection with the serum from the normal rabbits. The specific antibody was obtained in the immunized rabbits. The antibody titers of antiserum were measured by ELISA kits. Indirect competitive ELISA was used to determine serological specificity,and the obtained data was used to plot the inhibition curves. The content of Ig E antibody in the antiserum of rabbits was detected by rabbit immunoglobulin E(Ig E)ELISA kits.Results The antibody titer(OD)of isochlorogenic acid C was 2 times higher than that of the negative control,which indicated its potential allergenicity. The regression equation was I=0.430 lg C+0.319,with correlation coefficient of R2=0.997,and half inhibitory concentration as IC50=2.628 mg·mL-1. Furthermore,the exogenous Ig E antibody was produced in the rabbits. Conclusion The antiserum was prepared in rabbits after administration under the environmental conditions of 20 ℃ and 40% humidity in the laboratory. It was determined by ELISA that isochlorogenic acid C,a suspected hapten in Shuanghuanglian injection,was allergenic.
