高效液相色谱测定二十五味松生等丸中乌头碱的含量

【摘要】 旨在建立一种高效液相色谱法,用于测定藏药二十五味松生等丸中乌头碱的含量.对HPLC法的色谱条件进行了优化,流动相为甲醇-0.5%的三乙胺(70∶30),色谱柱为Aglient ZORBAX Eclipse Plus C18 (4.6 mm×250 mm, 5μm),检测波长为235 nm,流速为1.0 mL/min,进样量为10μL,柱温为室温.方法学研究结果表明,乌头碱的浓度在0.059μg/mL~3.330μg/mL范围内具有良好的线性关系(r=0.999 3),平均回收率为100.78%,RSD为1.16%.此方法操作简便、灵敏度高、结果准确可靠,可作为二十五味松生等丸中乌头碱的含量测定方法.

【Abstract】 To determine the content of aconitine in the Tibetan medicine Ershiwuwei Songshengdeng pills, an HPLC method was established. The chromatographic conditions of HPLC method were optimized: The determination was performed on an Aglient ZORBAX Eclipse Plus C18 column(4.6 mm × 250 mm, 5 μm) with mobile phase methanol-0.5% triethylamine(70∶30). The detection wavelength was set at 235 nm and the flow rate was 1.0 mL/min. The injection sample volume was 10 μL. The methodological study showed that the concentration of aconitine was linear in the range of 0.059 μg/mL to 3.330 μg/mL(r = 0.999 3). The average recovery was 100.78% with RSD of 1.16%. The method is simple, sensitive and accurate, and can be used as a method for the determination of aconitine in Ershiwuwei Songshengdeng pills.
