脱细胞神经联合骨髓间充质干细胞及富血小板凝胶治疗股神经损伤

【摘要】 背景:用异体脱细胞神经支架联合骨髓间充质干细胞和富血小板凝胶治疗兔股神经损伤,目前报道较少,机制并不明确。目的:评估脱细胞神经支架联合骨髓间充质干细胞和富血小板凝胶移植治疗兔股神经损伤的效果。方法:将25只新西兰兔随机分为5组,分别为单纯断端吻合组、脱细胞神经桥接组、脱细胞神经联合富血小板凝胶组、脱细胞神经联合骨髓间充质干细胞组、脱细胞神经联合富血小板凝胶和骨髓间充质干细胞组,术后12周称量腓肠肌湿质量,然后进行神经组织病理学染色。结果与结论:脱细胞神经联合富血小板凝胶和骨髓间充质干细胞组腓肠肌湿质量明显高于其他组(P <0.05),有较多的再生神经纤维生成,无炎性细胞浸润,神经丝蛋白NF-200阳性表达也较多,上述结果提示,脱细胞神经支架联合骨髓间充质干细胞和富血小板凝胶对于股神经损伤具有较好的修复效果。

【Abstract】 BACKGROUND: The use of allogeneic acellular nerve scaffold combined with bone marrow mesenchymal stem cells and platelet-rich gel to treat rabbit sciatic nerve injury is currently less reported and the mechanism is less clear.OBJECTIVE: To evaluate the effect of transplantation of acellular nerve scaffold combined with bone marrow mesenchymal stem cells and platelet-rich gel in the rabbit sciatic nerve injury model.METHODS: Twenty-five New Zealand rabbits were randomly divided into five groups: stump anastomosis group, acellular nerve scaffold group, acellular nerve scaffold combined with platelet-rich gel group, acellular nerve scaffold combined with bone marrow mesenchymal stem cell group, acellular nerve scaffold combined with platelet-rich plasma gel and bone marrow mesenchymal stem cell group. At 12 weeks after operation, the wet weight of gastrocnemius and histopathological staining experiments were performed. RESULTS AND CONCLUSION: The wet weight of gastrocnemius in the acellular nerve scaffold combined with platelet-rich gel and bone marrow mesenchymal stem cell group was significantly higher than that in the other groups(P < 0.05); moreover, there were many regenerated nerve fibers, no inflammatory cell infiltration, and positive expression of neurofilament protein NF-200. It is indicated that the combination of acellular nerve scaffold and bone marrow mesenchymal stem cells and platelet-rich gel has a good effect on the regeneration of peripheral nerve injury.
