山茱萸粗多糖的超声辅助提取及抗氧化活性研究

【摘要】 目的 制备山茱萸粗多糖并评价其抗氧化活性。方法 利用不同频率和功率的超声波辅助提取、制备山茱萸粗多糖,并进行抗氧化活性分析。结果 双频组合超声波浸提的效果优于单频超声波,最佳条件为超声频率28/45 kHz、超声功率330 W、料液比1∶6、浸提温度55℃,粗多糖的提取率可达19.15%。山茱萸粗多糖对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐[2,2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate),ABTS]的IC50值为0.26 mg·mL-1,最大清除率为65.71%,约为2,6-二叔丁基对甲酚(2,6-Di-tert-butyl-4-methylphenol, BHT)的2/3;山茱萸粗多糖对DPPH·的IC50值为0.28 mg·mL-1,最大清除率为95.70%,与BHT基本相同。结论 所得粗多糖具有抗氧化活性。

【Abstract】 Objective To prepare the crude polysaccharides of Cornus offcinalis and to evaluate the antioxidant activity. Methods The Cornus officinalis crude polysaccharides were prepared by ultrasonic assisted extraction with different frequency and power, and the antioxidant activity was analyzed. Results The results showed the extraction effect of dual frequency combined ultrasonic was better than that of single frequency ultrasonic. The optimal conditions were as follows: ultrasonic frequency 28/45 kHz, ultrasonic power 330 W, solid-liquid ratio 1∶6, and the extraction temperature 55 ℃. Under the above conditions, the extraction rate of crude polysaccharides could reach up to 19.15%. The IC50 value of the Cornus officinalis crude polysaccharides to 2, 2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS) was 0.26 mg·mL-1, and the maximum clearance rate was 65.71% which was about 2/3 of 2, 6-Di-tert-butyl-4-methylphenol(BHT). The IC50 value of Cornus officinalis crude polysaccharide for DPPH· was 0.28 mg·mL-1, and the maximum clearance rate was 95.70% which was basically the same as BHT. Conclusion The obtained crude polysaccharides have antioxidant activity.
