培养基优化对提高莫勒菌产孢量的影响
Effects of medium optimization on improving Moelleriella raciborskii sporulation
【摘要】 为提高昆虫病原真菌莫勒菌产孢量,通过正交试验设计L9(34),对莫勒菌培养基成分进行筛选和优化。结果表明,有助于莫勒菌产孢的碳源、氮源、金属离子和维生素分别为7 g·L-1可溶性淀粉、10 g·L-1酵母浸粉、4×10-4mol·L-1Fe2+和50 mg·L-1VC。莫勒菌在优化培养基上培养2周后的产孢量可达1.2×108孢子·cm-2,比优化前提高60倍。
【Abstract】 To improve sporulation,the composition of the growth medium for the entomopathogenic fungus Moelleriella raciborskii was optimized based on the orthogonal layout method including three levels of four variables.Specifically,7 g·L-1of soluble starch,10 g·L-1of yeast powder,4×10-4mol·L-1of ferrous ion,and 50 mg·L-1of ascorbic acid were the best sources of carbon,nitrogen,metal ions,and vitamins,respectively,for M.raciborskii sporulation.After 2 weeks of culture in this optimized medium,the spore production of M.raciborskii peaked,reaching1.2×108conidia·cm-2.Conidium production in the optimized medium was approximately 60-fold higher than that in the nonoptimized basal medium.
