甘肃野生羊肚菌根际土壤真菌群落与环境因子相互关系
Relationship between Fungal Community and Environmental Factors in Rhizosphere Soil around Wild Morels in Gansu
【摘要】 为探究甘肃不同采样点野生羊肚菌根际土壤中真菌群落结构以及真菌群落与环境因子的相关性,采用Illumina Mi-Seq高通量测序技术和生物信息学方法,分析真菌菌群组成和多样性,揭示真菌群落与环境因子的相互关系。结果表明,不同采样点的根际土壤中真菌群落差异较大;9个采样点共有的OTU(operational taxonomic unit)数为32个,特有的OTU数较多;迭部采样点羊肚菌根际土壤真菌多样性最高。所有采样点的优势菌门均为子囊菌门(Ascomycete)和担子菌门(Basidiomycota,),而纲、科、属水平优势类群各异。冗余分析(redundancy analysis, RDA)表明,子囊菌门与速效钾、谷氨酰胺酶、脲酶、硝态氮、碱性磷酸酶和脱氢酶箭头方向一致,说明子囊菌门与速效钾、谷氨酰胺酶、脲酶、硝态氮、碱性磷酸酶和脱氢酶正相关;Rozellomycota和Olpidiomycota与硒、铁、有效磷、氨态氮、脱氢酶呈正相关,与钙、多酚氧化酶、镁、pH呈负相关;壶菌门、unclassified_Fungi、Mortierellomycota与脱氢酶、碱性磷酸酶、硝态氮呈负相关。野生羊肚菌根际土壤真菌群落受酶活性影响较理化性质的影响大。
【Abstract】 This study aiming at exploring the reciprocity between fungal community structure as well as fungal community and environmental factors(FC-SEF) of wild morels in different sampling spots in Gansu Province. Illumina Mi-Seq high-throughput sequencing technology and informatics method were adopted to analyze the composition and diversity of fungal community, and reveal the reciprocity between fungal community and environmental factors. The results showed that there was wide difference among rhizosphere fungi from different sampling spots; there were a total of 32 OTU(operational taxonomic unit) from nine sampling spots, there were more peculiar OTU there; the diversity of morel rhizosphere soil fungi in overlapping sampling points was the highest. The dominant fungal phylum of all sampling spots was subphylum of ascomycetes and subphylum of basidiomycetes, and the dominant groups at class, family, and species level differ from each other. Redundancy analysis showed that the subphylum of ascomycetes accorded with the direction of arrow point of quick-acting potassium, glutaminase, urease, nitrate nitrogen, alkaline phosphatase, and dehydrogenase, showed that the subphylum of ascomycetes was positive correlation with quick-acting potassium, glutaminase, urease, nitrate nitrogen, alkaline phosphatase, and dehydrogenase. Rozellomycota and Olpidiomycota are positively correlated with Se, Fe content, available phosphorus, ammonia nitrogen, and dehydrogenase, but are negatively correlated with Ca, Mg content, pH value, and Peroxidase. Chytridiomycota, unclassified Fungi, and Mortierellomycota are negatively correlated with dehydrogenase, alkaline phosphatase and nitrate. Fungal communities in the rhizosphere soil of wild morels are more affected by enzyme activity than by physical and chemical factors.
