伊藤杂种‘巴茨拉’不同外植体无菌体系建立及愈伤组织诱导
Establishment of Aseptic System and Callus Induction of Paeonia Itoh ‘Bartzella’
【摘要】 选择‘巴茨拉’嫩叶、叶柄、茎段、鳞芽作为外植体,研究其无菌体系的建立及愈伤组织诱导的适宜激素和浓度及外植体形状对愈伤组织诱导过程的影响。结果表明:鳞芽外植体污染最为严重,其升汞处理适宜处理时间为12 min;茎段和叶柄为3 min,嫩叶为8 min。AC、PVP、Vc三种褐化抑制剂中PVP的褐化抑制效果最佳,适宜浓度为500 mg/L,其次是Vc,适宜浓度为1.5 mg/L,AC处理叶柄效果突出,适宜浓度为1.0 g/L。4种外植体中茎段和叶柄愈伤组织诱导效果理想,其适宜的激素为6-BA,浓度为1.0 mg/L。将外植体切成薄片状有利于缩短愈伤组织产生时间。
【Abstract】 The young leaf, petiole, stem segment and scale bud of ’bartzella’ were selected as explants to study the methods of inhibiting browning and pollution in the establishment of aseptic system, and to explore the best hormone type and concentration for callus induction. The results showed that the explants of scale bud were seriously polluted and were not suitable for callus induction.The optimal treatment time of HgCl2in stem segments and petioles was 3 min and that in young leaves was 8 min.Among the three b rowning inhibitors AC(activated carbon), PVP(polyvinylpyrrolidone) and VC(vitamin C), PVP has the best browning inhibition effect,with the use concentration of 500 mg/L, followed by VC, with the best concentration of 1.5 mg/l. The effect of AC treatment on petiole is outstanding, and the appropriate concentration is 1.0 g/L. Among the four explants, the callus induction effect of stem segment and petiole was ideal. The suitable hormone was 6-BA and the concentration was 1.0mg/l. Cutting explants into thin slices is helpful to shorten the time of callus production.
