水冬瓜叶药材的质量标准研究

【摘要】 目的:建立水冬瓜叶的质量标准。方法:参照《中国药典》(2015年版)四部项下药材和饮片检定通则要求及相关项目检测方法,测定水分、总灰分、浸出物含有量;采用薄层色谱法(TLC),以乙酸乙酯-甲酸-水(33∶4∶2)为展开系统定性鉴别水冬瓜叶药材;采用高效液相色谱法(HPLC),以0.2%磷酸水-乙腈为流动相,Agela Promosil C18为色谱柱测定金丝桃苷含量。结果:建立的薄层色谱中金丝桃苷斑点清晰,分离度高,重复性好。10批水冬瓜叶药材水分、总灰分、浸出物含有量分别为6.34%~8.89%、7.95%~11.62%、29.48%~39.76%。金丝桃苷在0.008 24~0.164 80 mg/ml(r=0.999 2)范围内线性关系良好;精密度、稳定性、重复性的RSD均小于3.0%;平均加样回收率为100.53%,RSD为2.67%(n=6)。结论:初步拟定水冬瓜叶药材中水分不得超过10.3%,总灰分不得超过12.0%,浸出物不得少于25.0%,金丝桃苷含量以干燥品计不得少于0.051%。建立的方法稳定可行,专属性强,可用于水冬瓜叶的质量控制。

【Abstract】 Objective:To establish the quality standard for the leaves of Toricellia Angulata Oliv.var.intermedia(Harms)Hu. Methods:The contents of moisture,total ash and extract were determined according to the requirements of the general rules for the verification of medicinal materials and decoction pieces and the detection methods of related items under the four parts of the Chinese Pharmacopoeia(2015 Edition). Thin layer chromatography(TLC) was used to qualitatively identify the Toricellia Angulata Oliv.var.intermedia(Harms)Hu leaves with ethyl acetate-formic acid-water(33∶4∶2) as the development system. The content of hyperin was determined by HPLC with 0.2% phosphoric acid-acetonitrile as the mobile phase and Agela Promosil C18 as the chromatographic column. Results:The spots of hyperin in the established TLC were clear,with high resolution and good repeatability. The contents of 10 batches of the leaves of Toricellia Angulata Oliv.var.intermedia(Harms)Hu of water,total ash and extract were 6.34%~8.89%,7.95%~11.62%,and 29.48%~39.76%,respectively.The linear range of hyperin was 0.008 24~0.164 80 mg/ml(r = 0.999 2).RSDs of precision,stability,and repeatability were all less than 3.0%. The average recovery of hyperin was 100.53%(RSD was 2.67%,n = 6). Conclusion:It is suggested that the moisture and total ash should not exceed 10.3% and 12.0%,respectively. The contents of extract content should not be less than 25.0%. The content of hyperin in dried products should not be less than 0.051%. The established quality standards for the leaves of Toricellia Angulata Oliv.var.intermedia(Harms)Hu.is stable,feasible,and highly specific and can be effectively used as the quality control for the leaves of Toricellia Angulata Oliv.var.intermedia(Harms)Hu.
