玻璃化冷冻对卵母细胞、胚胎和子代的影响及其相关表观遗传学的研究进展

【摘要】 玻璃化冷冻技术目前已逐步取代程序化冷冻技术并广泛运用于体外受精-胚胎移植助孕过程。虽然卵母细胞及胚胎经玻璃化冷冻后有较好的临床结局,但玻璃化冷冻过程中使用的高浓度冷冻保护剂、体外冷冻复苏等操作是否对卵母细胞、胚胎以及其子代的安全性造成不良影响仍然备受关注。分子层面研究结果表明,虽然玻璃化冷冻不改变哺乳动物DNA序列,但可导致基因表观遗传修饰水平的变化进而影响其靶基因表达水平。本文从哺乳动物卵母细胞和胚胎经玻璃化冷冻后对胚胎、妊娠结局和子代的影响以及对相关表观遗传修饰水平的改变进行综述,探索玻璃化冷冻对卵母细胞和胚胎影响的分子机制,为进一步提高卵母细胞和胚胎的冷冻保存技术提供理论依据。

【Abstract】 Vitrification technology has gradually replaced programmed freezing technology and has been widely used in IVF-ET. Although oocyte and embryo have good clinical outcome after vitrification, whether the high concentration cryoprotectants used in vitrification and frozen-thawed process have adverse effects on the safety of oocytes, embryos and their offspring is still of great concern. The molecular level study showed that although vitrification did not change mammalian DNA sequence, it could lead to changes in the level of epigenetic modification of genes and thus affect the level of target gene expression. This paper reviews the effect of vitrification of mammalian oocyte and embryo on embryo, pregnancy outcome and offspring, as well as changes in the level of related epigenetic modifications, and explores the molecular mechanism of the effects of vitrification on oocytes and embryos, in order to provide a theoretical basis for further improving the cryopreservation technology of oocyte and embryo.
