【摘要】 目的 探讨发酵桦褐孔菌(FIO)诱导人肝癌细胞HepG-2凋亡的作用及机制。方法 MTT法测定细胞增殖,HE染色法观察细胞形态,流式细胞仪检测细胞凋亡及细胞周期,蛋白免疫印迹法检测细胞内的凋亡相关蛋白的表达。结果 FIO显著抑制HepG-2细胞的增殖,且具有时间和剂量相关性;FIO作用于HepG-2细胞后出现明显凋亡形态、凋亡率显著增加且呈浓度依赖性,细胞周期明显阻滞在G0/G1期;FIO各剂量组细胞内Caspase-3,Caspase-9,Cyt-c, Bax/Bcl-2和p53蛋白表达均显著升高,而ProCaspase-3和ProCaspase-9蛋白表达显著减少,且其作用呈现浓度依赖性。结论 FIO对HepG-2细胞具有抑制增殖和诱导凋亡的作用,而线粒体通路可能参与调节FIO诱导HepG-2细胞凋亡的作用。

【Abstract】 Objective To investigate the apoptotic effect of fermented Inonotus Obliquus(FIO) on human hepatocellular carcinoma HepG-2 cells. Methods MTT assay was used to determine the cell proliferation, and the cell morphology was observed by HE staining. Flow cytometry was used to determine the apoptosis and cell cycle of the cells, and the expressions of apoptosis-associated proteins in HepG-2 cells were detected by Western blotting. Results FIO significantly inhibited the proliferation of HepG-2 cells and which correlated with the time and the concentration of administration. After treat with FIO, HepG-2 cells were appeared the apoptotic morphology, the apoptosis rate of the cell was significantly increased in a concentration dependent manner and the cell cycle of the cell was arrested at G0/G1 phase. The expressions of Caspase-3, Caspase-9, Cyt-c, Bax/Bcl-2 and p53 proteins were significantly increased and the expressions of ProCaspase-3 and ProCaspase-9 proteins were significantly decreased in the FIO groups.Conclusion FIO inhibits the proliferation and induces the apoptosis of HepG-2 cells, and mitochondrial pathway may participate in the FIO induced apoptosis of HepG-2 cells.
