地黄中环烯醚萜苷类成分在根际土壤中的分布特征研究
Distribution characteristics of iridoid glycosides in rhizosphere soil from Rehmannia glutinosa
【摘要】 目的 研究不同种质地黄生长期内植株中苦味物质环烯醚萜苷类成分在根际土壤中的分布特征,为揭示地黄连作障碍的成因提供理论依据。方法 以BJ-1、85-5、金九、BX 4个不同种质的地黄为研究对象,采用药典中梓醇含量测定方法与建立的HPLC方法分别测定四个地黄种质生长期内块根、叶片与根际土壤中多种环烯醚萜苷成分的含量,分析其积累及分布特征。结果 生长期内不同种质地黄块根和叶片中均检测出梓醇、地黄苷D、地黄苷A和益母草苷4种环烯醚萜苷类成分,根际土壤中检测出梓醇、地黄苷D和益母草苷3种环烯醚萜苷类成分;梓醇和益母草苷为生长期内地黄块根和叶片中含量较高的环烯醚萜苷,而根际土壤中含量较高的为地黄苷D;不同种质地黄根际土壤中环烯醚萜苷类物质的变化趋势基本一致,但含量存在差异。结论 地黄根系分泌的环烯醚萜苷类物质可以在地黄根际土壤中检测到,但生长期内地黄根际土壤中的环烯醚萜苷类物质的分布特征与地黄植株无明显相关性;根际土壤中检测到的环烯醚萜苷是否是种植地黄后引起地黄土壤变苦的主要物质及根际土壤中的环烯醚萜苷与地黄连作障碍形成的关系还有待于进一步研究。
【Abstract】 Objective The distribution characteristics of bitter substances iridoid glycosides in different germplasm rhizosphere soil during the growth period of Rehmannia glutinosa were studied to provide theoretical basis for revealing the causes of continuous cropping obstacles of R. glutinosa. Methods The contents of iridoid glycosides in root tuber, leaf and rhizosphere soil of four germplasms of R. glutinosa(BJ-1, 85-5, Jinjiu and BX) in growth period were determined by the determination method of catalpol in pharmacopoeia and the established HPLC method, analyzing the accumulation and distribution characteristics.Results Four iridoid glycosides including catalpol, rehmannia glycoside D, rehmannia glycoside A and leonurus glycoside were detected in root tuber and leaves of different germplasm R. glutinosa during growth period, and three iridoid glycosides including catalpol, rehmannia glycoside D and leonurus glycoside were detected in rhizosphere soil. Catalpa and leonurus glycoside were the iridoid glycoside with higher content in root tuber and leaf of R. glutinosa, while Rehmannia glutinin D was the one with higher content in rhizosphere soil. The changing trend of iridoid glycosides in rhizosphere soil of R. glutinosa from different germplasm was mainly the same, but the contents of iridoid glycosides were distinct. Conclusion R. glutinosa plants could release iridoid glycosides into the rhizosphere soil during the growth period, but there was no significant correlation between the accumulation characteristics of iridoid glycosides in the rhizosphere soil and the plants. Whether the bitter substances of iridoid glycosides released from the rhizosphere soil during the growth period of R. glutinosa are the main chemical components causing the soil bitter remains to be further studied.
