桂枝茯苓丸混合粉末质量快速分析方法研究

【摘要】 目的 基于高效液相色谱、近红外光谱(near infrared spectroscopy, NIRS)及多变量数据处理技术,提出了一种桂枝茯苓丸混合粉末质量快速分析方法,并用于预测桂枝茯苓丸混合粉末中肉桂酸和丹皮酚的含量。方法 以高效液相色谱法作为肉桂酸和丹皮酚含量测定的参照方法,使用偏最小二乘回归法建立桂枝茯苓丸混合粉末近红外光谱与肉桂酸、丹皮酚含量之间的多元校正模型,并以15批外部验证集样本对模型性能进行评价。结果 偏最小二乘回归法建立的模型相关系数可达0.99,外部验证集的相关系数也高于0.92。结论 该方法准确、稳健、快速,适用于桂枝茯苓丸混合粉末质量的快速评价。

【Abstract】 Objective A rapid analysis method based on high performance liquid chromatography(HPLC), near infrared spectroscopy(NIRS) and multivariable data analysis was established for quality evaluation of Guizhifulingwan Mixture. The contents of active ingredients of Guizhifulingwan Mixture including cinnamic acid and paeonol were simultaneously detected by using this method.Methods NIRS HPLC was used as a reference method for determining the contents of cinnamic acid and paeonol. Partial least squares(PLS) analysis was implemented as a linear method for multivariate models calibrated between NIR spectrum and contents of cinnamic acid and paeonol. Finally, the performance of the model was evaluated by 15 batches of test samples. Results The results showed that NIR models gave a good calibration ability with an R2 value above 0.99, and the prediction ability was also satisfactory with an R2 value higher than 0.92. Conclusion The overall results demonstrate that the established method is accurate, robust and fast, therefore, it can be used for rapid quality evaluation of Guizhifulingwan Mixture.
