黑素瘤缺乏因子2和线粒体DNA在银屑病皮损和血清中的表达水平测定
Detection of AIM2 in skin lesions and mtDNA in serum of psoriasis patients
【摘要】 目的 检测寻常性银屑病患者皮损组织中黑素瘤缺乏因子2(AIM2)表达水平和血清中线粒体DNA(mtDNA)表达水平;观察poly(dA:dT)转染HaCaT细胞对HaCaT细胞增殖以及AIM2表达的影响,初步探讨AIM2和mtDNA在银屑病发病机制中的作用。方法 实时荧光定量聚合酶链反应(qRT-PCR)检测寻常性银屑病患者组(PSO组)与健康对照组(CON组)血清中mt DNA mRNA表达水平;Western印迹(Western-blot)检测PSO和CON组皮肤组织中AIM2蛋白水平的表达差异;CCK-8法检测不同浓度的poly(dA:dT)转染HaCaT细胞增殖的情况;qRT-PCR法检测各组细胞中AIM2 mRNA的表达情况。结果 q RT-PCR检测示PSO组血清中mt DNA mRNA表达水平明显升高,与CON组相比差异有统计学意义(P <0.000 1);Western印迹示,PSO组皮损中AIM2蛋白表达水平明显升高,与CON组相比差异有统计学意义(P<0.001);细胞CCK8增殖实验示2μg/ml poly(dA:dT)及5μg/ml poly(dA:dT)处理组细胞增殖能力升高,其中5μg/ml poly(dA:dT)处理组细胞增殖能力提升明显,且差异有统计学意义(P <0.001);细胞qRT-PCR检测示2μg/ml及5μg/ml poly(dA:dT)处理组AIM2 mRNA的表达水平较对照组升高,且差异具有统计学意义(P <0.05,P <0.000 1)。结论 AIM2在银屑病患者皮损中高表达,AIM2内源性诱导因子mtDNA在银屑病患者血清中升高,且体外试验诱导AIM2升高可促进HaCaT细胞增殖,或提示AIM2可能参与了银屑病的发病。
【Abstract】 Objective To detect the expression levels of absent in melanoma 2(AIM2) in the skin lesions and Mitochondrial DNA(mt DNA) in serum of patients with psoriasis vulgaris. To observe the effect of the proliferation and AIM2 expression of HaCaT cells transfected with poly(dA:dT), and to preliminarily explore the role of AIM2 and mtDNA in the pathogenesis of psoriasis. Methods Quantitative real timepolymerase chain reaction(qRT-PCR) method was used to detect the expression level of mtDNA mRNA in the serum of the psoriasis vulgaris group(PSO group) and the control group(CON group). The expression of AIM2 protein in skin tissues of PSO group and CON group was detected by Western-blot. Cell Counting Kit-8(CCK-8) assay was used to detect the proliferation of HaCaT cells transfered by poly(dA:dT) of different concentrations. The expression levels of AIM2 mRNA in each group were detected by qRT-PCR. Results The qRT-PCR results showed that the expression levels of mtDNA mRNA in serum of PSO group patients were significantly higher than the CON group, and the difference between groups was statistically significant(P<0.000 1). Western blot analysis showed that AIM2 protein expression level in skin lesions of psoriasis patients was significantly higher than the CON group, and the difference between groups was statistical significant(P<0.001). CCK8 cell viability assay showed that cell proliferation ability of 2μg/ml poly(dA:dT) treatment group and 5 μg/ml poly(dA:dT) treatment group was increased. Compared with untreated group, cell proliferation ability of 5 μg/ml poly(dA:dT) treatment group was significantly improved, and the difference was statistically significant(P<0.001). qRT-PCR results showed that the expression levels of AIM2 mRNA in 2μg/ml Poly(dA:dT) group and 5μg/ml poly(dA:dT) group were higher than that in untreated group,and the difference was statistically significant(P<0.05, P<0.000 1, respectively). Conclusion AIM2 is highly expressed in the skin lesions of patients with psoriasis. mtDNA is elevated in the serum of patients with psoriasis vulgaris. AIM2 can promote the proliferation of HaCaT cells in vitro. Above all, our results suggest that AIM2 may be involved in the pathogenesis of psoriasis.
