不饱和脂肪酸对肝癌移植瘤小鼠P21、P53、P13K及PKB/Akt蛋白表达的影响
Effects of Unsaturated Fatty Acids on the Expression of P21, P53, P13K and PKB/Akt in Liver Cancer Transplanted Mice
【摘要】 目的 建立SCID小鼠肿瘤模型,为筛选新的抗肿瘤药物提供受试对象。方法 将肝癌组织移植到重症联合免疫缺陷(SCID)小鼠皮下,待移植瘤生长到6~7 mm时,50只荷瘤SCID小鼠分为不饱和脂肪酸低、中、高剂量组【50 mg/(kg·d)、100/(kg·d)、200 mg/(kg·d)】,五氟尿嘧啶(5-FU)组【100 mg/(kg·d)】,模型组【蒸馏水2 mL/(kg·d)】,连续给药21 d,剥取移植瘤测量其体质量和体积,比较肿瘤生长情况;用酶联免疫吸附法(ELISA)检测瘤组织P21、P53、P13K、PKB/Akt蛋白表达水平。结果 不饱和脂肪酸低、中、高剂量组和5-FU组肝癌移植瘤质量或体积显著小于模型组(P<0.01);肝癌移植瘤P21水平较模型组明显升高(P<0.05,P<0.01);P53、P13K、PKB/Akt水平较模型组明显降低(P<0.05,P<0.01)。结论 不饱和脂肪酸对肝癌移植瘤生长具有明显的抑制作用。其可能机制与上调移植瘤P21水平,下调P53、P13K、PKB/Akt水平,抑制PI3K-Akt信号传导通路活性有关。
【Abstract】 Objective To establish SCID mice tumor model for screening new antitumor drugs.Method Liver cancer tissue was transplanted subcutaneously into severe combined immune deficiency(SCID) mice. When the transplanted tumor grew to 6-7 mm, fifty SCID tumor-bearing mice were divided into unsaturated fatty acid low, medium and high dose groups [50 mg/(kg·d), 100/(kg·d), 200 mg/(kg·d)]. Pentafluorouracil(5-FU) group [100 mg/(kg·d)] and model group [distilled water 2 mL/(kg·d)] were administered continuously for 21 d. The transplanted tumor was removed to measure its quality and volume, and the tumor growth was compared. The protein expression levels of P21, P53, P13 K and PKB/Akt in tumor tissues were determined by enzyme-linked immunosorbent assay(ELISA).Result The weight or volume of liver cancer transplanted tumor in low, medium and high dose of unsaturated fatty acids groups and 5-FU group was significantly lower than that in model group(P<0.01). The level of P21 was significantly higher compared with model group(P<0.05, P<0.01),the levels of P53, P13 K and PKB/Akt were significantly lower than those in model group(P<0.05, P<0.01).Conclusion Unsaturated fatty acids can inhibit the growth of liver cancer transplanted tumor, The possible mechanism is related to up-regulation of P21 level, down-regulation of P53, P13 K, PKB/Akt levels, and inhibition of PI3 K-Akt signal transduction pathway activity.
